Yellow Sea surface sediment samples collected on October 15-31, 2003 were analyzed using the palynological process to investigate the spatial distribution of dinoflagellate resting cysts. The sampling areas comprised four latitudinal transects, the northernmost of which was located off the Shandong Peninsula, China and the southernmost off Jeju Island, Korea. Each transect line was composed of six to nine stations, spanning the distance between the Chinese and Korean coasts. Twenty-five different types of dinoflagellate cysts were identified. Gonyaulax scrippsae, Alexandrium spp. (ellipsoidal type), and G. spinifera were the most dominant at all stations surveyed. Dinoflagellate cysts belonging to the Gonyaulacales comprised over 50% of all cysts collected. The latitudinal distribution trend showed that cyst concentrations along the two middle transects were much higher than those along the two northern and southern transects. Cyst concentrations in the offshore central areas reached their highest values within each transect and gradually decreased toward the Chinese and Korean coasts. Overall, cyst concentrations were markedly elevated in the offshore central Yellow Sea areas and gradually decreased outward in all four directions. This concentric cyst distribution pattern was consistent with the hydrographic features of the Yellow Sea, such as circular current systems, sedimentary properties, and water depth.

The total dinoflagellate cyst community and the cysts of Cochlodinium polykrikoides in the surface sediments of South Sea (Tongyeong coast), South Korea, were analysed using next-generation sequencing (NGS) and morphological approaches. Dinoflagellate cysts can be highly abundant (111–4,087 cysts g−1 dry weight) and have diverse species composition. A total of 35 taxa of dinoflagellate cysts representing 16 genera, 21 species (including four unconfirmed species), and 14 complex species were identified by NGS analysis. Cysts of Scrippsiella spp (mostly Scrippsiella trochoidea) were the most dominant and Polykrikos schwartzii, Pentapharsodinium dalei, Ensiculifera carinata, and Alexandrium catenella/tamarense were common. Thus, a combination of NGS and morphological analysis is effective for studying the cyst communities present in a given environment. Although C. polykrikoides developed massive blooms during 2013–2014, microscopy revealed low density of their cysts, whereas no cysts were detected by NGS. However, the vegetative C. polykrikoides not appeared during 2015–2017 in spite of the observation of C. polykrikoides cysts. This suggests that the C. polykrikoides blooms were not due to development of their cysts but to other factors such as currents transporting them to a marine environment suitable for their growth.

Resting cysts of the marine phytoplanktonic dinoflagellate Scrippsiella spp. are encountered in coastal habitats and shallow seas all over the world. Identification of Scrippsiella species requires information on cyst morphology because the plate pattern of the flagellated cell is conserved. Cysts from sediments of the East China Sea were identified based on traits from both the cysts and the thecal patterns of germinated cells. Calcareous cysts belonged predominantly to S. trochoidea (F. Stein) A. R. Loebl., S. rotunda J. Lewis, and S. precaria Montresor et Zingone. The former two species also produced smooth and noncalcified cysts in the field. A new species, S. donghaienis H. Gu sp. nov, was obtained from six noncalcified cysts with organic spines. These cysts are spherical, full of pale white and greenish granules with a mesoepicystal archeopyle. The vegetative cells consist of a conical epitheca and a round hypotheca with a plate formula of po, x, 4', 3a, 7'', 6c (5c + t), 6 s, 5''', 2'''' and are morphologically indistinguishable from S. trochoidea. Results of internal transcribed spacer (ITS) sequence comparisons revealed that S. donghaienis was distinct from the S. trochoidea complex and appeared nested within the Calciodinellum/Calcigonellum clade. Culture experiments showed that the presence of a red body in the cyst and the shape of the archeopyle were constant within cell lines from one generation to the next, while the morphological features of the cyst wall, such as calcification and spine shape, appeared to be phenotypically plastic.
