Dimethyleacetamide improves the cryosurvivability of Indian red jungle fowl ( Gallus gallus murghi ) sperm

Abstract: It was hypothesized that dimethyleacetamide (DMA) can be used as an alternate to glycerol for cryopreservation of Indian red jungle fowl semen. Four concentrations of DMA (4%, 6%, 8% and 10%) in extender were compared with previously optimized cryopreservation protocol based on 20% glycerol (control) for Indian red jungle fowl. Sperm motility, plasma membrane integrity, viability, and acrosome integrity were assessed at the stage of post-dilution, cooling, equilibration, and freeze-thawing. The whole experimen… Show more

“…The presence of protein denaturation and changes in actin interactions within the spermatozoa tail may interfere with motility and, consequently, adversely affect fertilization (Shaliutina, Hulak, Gazo, Linhartova, & Linhart, ; Viveiros et al, ). Similar results were obtained for equine semen (Pace & Sullivan, ) and in Indian red jungle fowl (Blesbois et al, ; Rakha et al, , ).…”

“…These inconsistent results might be explained by species-specific differences in the membrane composition and seminal plasma content. In domestic bird species, DMA is often recommended for sperm cryopreservation in pellets, and it yields better results when applied to protocols with rapid cooling rates [15,20,23]. In contrast, DMSO is better suited to slow freezing methods [24].…”

“…In contrast, DMSO is better suited to slow freezing methods [24]. Additionally, low fertility outcomes have been observed when DMA/straw-based cryopreservation was compared with sperm cryopreserved with glycerol; this finding has been attributed to a negative interaction between the plastic straws and the cryoprotectant [23].…”

“…Other methods including primordial germ cells and gonadic tissues technologies are still in progress but may be seen as methods that are complementary to semen cryopreservation (Liptoi et al., 2013; Nakamura, 2016; Sztan et al., 2017; Thélie et al., 2018). Different freezing procedures have been developed to cryopreserve avian semen (Blesbois et al., 2007; Santiago-Moreno et al., 2012; Rakha et al., 2016, 2017a; 2018a,b,c; Thananurak et al., 2016; Chuaychu-noo et al., 2017; Thananurak et al., 2017). However, simplification of the more efficient common methods of chicken sperm cryopreservation is still needed in order to make them available to large environmental and breeding conditions.…”
