Dimethyl sulphoxide dose–response on rat retinal function

Abstract: The aim of the study is to determine whether dimethyl sulphoxide (DMSO), a common laboratory solvent, impacts retinal function. Long Evans rats (n = 17) were intravitreally injected with five different doses of DMSO representative of those reported in the literature and spanning over 3 log units of vitreal concentration (0.01-8%). Retinal function was evaluated 1 h after injection using electroretinograms, and the waveform was decomposed into outer (photoreceptor), middle (ON-bipolar cell) and inner retinal (a… Show more

“…5, DMSO‐induced cell death increased expression of Bax in mitochondrial extracts at final concentrations of 2 and 4 but not 1% (v/v), up to 24 h after exposure. This observation is in agreement with the existing literature where 1% DMSO treatment for up to 40 h in U937 cells (the human myeloid leukemia cell line) did not show changes in Bax levels (46). Likewise, in an in vivo model, Dalton's lymphoma mice mRNA levels were assessed and the levels of Bax increased with the concomitant decrease of the Bcl‐2/ Bax ratio after DMSO treatment (7.5 g/kg ip; ref.…”

“…16, 18), but most often, its absolute concentration is not reported (8, 9, 1517). There is some functional, although no structural evidence that retinal toxicity is induced by low concentrations (0.6–8% v/v) of DMSO, but this study was based only on electrophysiology data (46).…”

“…Apigenin was administrated intraperitoneally dissolved in dimethyl sulfoxide solvent (DMSO). [2526] The exact consistency of the solution was: 5 mg apigenin + 0.2 ml DMSO + 0.3 ml NaCl 0.9%. Reperfusion was followed for 60, 120, and 240 minutes (AP60, AP120, and AP240 groups).…”

“…APB and TPMPA were diluted using distilled H 2 O, whereas CNQX was mixed in the minimal volume of dimethyl sulphoxide (DMSO) that gave dilution. This DMSO concentration is known to have little effect on retinal function [25]. The concentrations stated represent the estimated final vitreal concentrations assuming complete dilution and a rat vitreal volume of 50  μ L [26].…”
