Diglons are heterodimeric proteins composed of IgLON subunits, and Diglon-CO inhibits neurite outgrowth from cerebellar granule cells

Abstract: IgLONs are a family of four cell adhesion molecules belonging to the Ig superfamily that are thought to play a role in cell-cell recognition and growth-cone migration. One member of the family, opioid-binding cell-adhesion molecule (OBCAM), might act as a tumour suppressor. Previous work has shown that limbic-system-associated protein (LAMP), CEPU-1/Neurotrimin and OBCAM interact homophilically and heterophilically within the family. Here, we show that, based on their relative affinities, CEPU-1 might be both … Show more

“…It is novel in various CAMs that homophilic and heterophilic cis-interactions are important for trans-interactions to control neuronal outgrowth and synaptogenesis. Four IgLON CAMs are able to form six heterophilic dimer called Diglons, 16 therefore, it is possible that homophilic and heterophilic cis-interactions of IgLON proteins at postsynaptic sites are crucial to modulate the synaptic formation through presynaptic partners of IgLON proteins. Data represent means AE s.e.m obtained from 13 control (total analyzed length; 2050 mm), 9 Kilon-expressing (1700 mm), 19 LAMP-expressing (3200 mm), 9 Ntm-expressing (1500 mm), 12 OBCAM-expressing (1400 mm), 12 Kilon-LAMP-expressing (2150 mm), 11 Kilon-Ntm-expressing (1950 mm), 11 Kilon-OBCAM-expressing (1850 mm), 13 LAMP-Ntmexpressing (2600 mm), 10 LAMP-OBCAM-expressing (1700 mm), and 12 Kilon-expressing neurons (2600 mm).…”
