Digital Image Analysis of Platelet-Extracellular Matrix Interactions: Studies in von Willebrand Disease Patients and Aspirin-Treated Donors

Abstract: We have developed an automated image analysis method which provides en face morphometric evaluation of platelet interactions with coverslips coated with extracellular matrices. This procedure gives also additional information on the intensity of platelet-surface interactions in the same specimens. To standardize the method, anticoagulated human blood was flowed at 800 s-1 during 10 min at 37°C through a parallel perfusion chamber in which extracellular matrix (ECM)-coated coverslips were placed. The… Show more

“…A software package (SigmaScan image analysis software, version 4.00.023; Jandel Scientific-Systat, Chicago, IL) provides a specialized program to determine the gray level histogram of each image. 34 Through successive mathematical calculations, this computer program determines the percentage of surface covered by platelets (% P) versus the total area screened.…”

“…The percentage of vWF activity is obtained from aggregometer tracings. 12 vWF collagen binding activity This assay was performed according to a previously described method 10 with minor modifications. Plasma samples diluted 1:100 with PBS containing 0.2% bovine serum albumin were placed on multiwell plates precoated with collagen type III (5 µg/ml) and blocked with 1% bovine serum albumin in PBS for 15 minutes.…”

“…Number and degree of spreading of adhered platelets were evaluated in 10 random microscope fields per coverslip by an automatic method. 11 The mean number of platelets adhered on the coverslip was measured and expressed per 1000 mm 2 . The degree of platelet activation was classified in four categories, according to previously established criteria, 12 as follows: type 0 (contact; round platelets in contact without pseudopods); type I (early dendritic; platelets in early stages of spreading, with several pseudopods extending radially); type II (late dendritic; platelets in a more advanced state of activation with cytoplasms beginning to fill the spaces between pseudopods); and type III (fully spread; the cytoplasms of adhered platelets filling completely the spaces between pseudopods, in a final activation state).…”
