Differentiation of Murine NK Cells into Distinct Subsets Based on Variable Expression of the IL-12Rβ2 Subunit

Abstract: The cytokine IL-12 manifests its biological activity via interaction with a heterodimeric receptor (IL-12R) present on activated T and NK cells. The cDNAs for two IL-12R subunits have been cloned from human and mouse and designated IL-12Rβ1 and IL-12Rβ2. The expression of IL-12Rβ2 on T cells is influenced by cytokines, particularly IL-4, IL-12, and IFN-γ; however, little is known regarding regulation of IL-12R expression on NK cells. In this study we show that murine NK cells differentiate into IL-12Rβ2low and… Show more

“…33 Previous reports suggested that mouse NK cells might differentiate in a manner similar to human NK cells, 12,26 and a NK1-NK2 paradigm was proposed for both human and mouse NK cells. 34,35 It was proposed that immature human NK cells produced IL-13 but not IFN-␥, and thus were defined as type 2 NK cells expressing TRAIL. 15,[36][37][38] Differentiation to an intermediate stage of human NK cells (defined as type 0) led to both IL-13 and IFN-␥ production and the expression of TRAIL, FasL, and perforin.…”

“…Since IL-12R␤1 is shared with IL-23 (33) and both heterodimer chains (IL-12R␤1 and IL-12R␤2) are required to produce high affinity to IL-12, it is likely that up-regulation of IL-12R␤2 will induce a specific increase in affinity to IL-12. This notion was supported by findings in a mouse model, where IL-12R␤2-deficient mice were defective in IL-12-mediated signaling (10,54). Our results, combined with those of others (10,53), show that although IL-12R␤1 is the subunit primarily responsible for the binding of IL-12, IL-12R␤2 plays an essential role in mediating the biological functions of IL-12.…”

“…In addition, freshly separated NK cells from chronic hepatitis C virusinfected patients showed significant production of IL-10 and normal concentrations of IFN-␥ upon cell-mediated triggering (41). Evidence for the presence of these NK cell subsets was also supported in mice, which showed that mouse IL-2-activated NK cells can be subdivided based on expression of high or low levels of the IL-12R␤2 (42). It has been reported that populations of peripheral blood IL-10-producing (NKr1)-CD56 bright and CD56 dim NK cells in early pregnancy women were increased compared with spontaneous abortion cases (43).…”
