Differentiation‐dependent induction of CYP1A1 in cultured rat small intestinal epithelial cells, colonocytes, and human colon carcinoma cells: Basement membrane‐mediated apoptosis

Abstract: Rat small intestinal epithelial cells and human colon adenocarcinoma cells cultured on Matrigel expressed the differentiation specific enzyme, sucrase-isomaltase, as determined by indirect immunofluorescence. Rat small intestinal epithelial cells, rat colonocytes, and human colon adenocarcinoma cells developed an altered morphology when cultured on Matrigel and became apoptotic within 24-48 h. Benzo[a]pyrene and 2,3,7,8-tetrachlorodibenzo-p-dioxin caused a 2- and 5-fold induction, respectively, of ethoxyresoru… Show more

“…The TGF receptors are intimately related with MAP-kinases mediated apoptotic pathway (JNK, p53/ Erk), whereas p53 is activated in the endpoints of the pathway [44][45][46]. The growth-inhibitory apoptotic potential and growth factor-modulator effects of maslinic acid could be related with MAP-kinases mediated apoptotic pathway [22]. Other MAPkinases pathways as the TNF1a receptor-JNK-jBID pathway could be related with apoptotic activation by maslinic acid [47].…”

“…All these features are consistent with those of intestinal crypt cells. Since the establishment and characterization of this cell line, IEC-6 cells have been used in numerous studies to elucidate mechanisms of differentiation [21,22].…”

“…Moreover, EROD activity in Caco-2 cells cultured on plastic was compared with that of cells cultured on Matrigel, with or without the CYP1A1 inducers benzo[a]pyrene and 2,3,7,8-tetrachlorodibenzo-p-dioxin (Sterling and Cutroneo, 2002). In both culture conditions, benzo[a]pyrene increased EROD activity by about 7-fold compared to non-induced cells, while 2,3,7,8-tetrachlorodibenzo-p-dioxin induction was 2-fold greater on Matrigel-grown cultures than on plastic-grown cells.…”

“…CYP1A1 (Zhang et al 1996;Spatznegger, Horsmans, and Verbeck 2000;Sterling and Cutroneo 2002;Roos 2002) and UDP-glucuronyltransferases (Dubey and Singh 1988) that are involved in the metabolism of PAHs have been isolated from the small intestine. Ingestion of PAHs causes a very rapid and profound autoinduction of intestinal CYP1A1 that occurs as a consequence of the kinetics of intestinal absorption.…”
