Differential staining of peripheral nuclear chromatin with Acridine orange implies an A-form epichromatin conformation of the DNA

Abstract: The chromatin observed by conventional electron microscopy under the nuclear envelope constitutes a single layer of dense 30–35 nm granules, while ∼30 nm fibrils laterally attached to them, form large patches of lamin-associated domains (LADs). This particular surface “epichromatin” can be discerned by specific (H2A+H2B+DNA) conformational antibody at the inner nuclear envelope and around mitotic chromosomes. In order to differentiate the DNA conformation of the peripheral chromatin we applied an Acridine oran… Show more

“…Similarly, the preferential way of the chromatin networking might be determined by the anisotropic (symmetry breaking) super-packaging of heterochromatin [54], with its aggregation-stimulating capacity to impinge 'a sticky silence' on the large genome domains [15,55]. The non-coding RNA that is transcribed from heterochromatin [56] can also electrostatically contribute to its packaging [25,57]. The spatial compartmentalization of the nucleus is maintained by relatively simple basic physicochemical principles, including electrostatic and hydrophobic forces that generate an extremely detailed 'nuclear topology' giving a material basis to robust gene expression regulation, as indicated by Ronald Hancock [58].…”

“…Epichromatin is a conformational epitope formed by DNA and histones H2A and H2B localized only at the exterior chromatin surface [17,18]. More recently, it was observed that epichromatin forms superbead domains associated to DNA-A at the nuclear envelope [19]. A 3D analysis also shows that P. falciparum nucleus presents a polarization of the nuclear pore complex: in the early multinucleated schizont, it clusters in the nucleus region facing the mother plasma membrane, whereas in the late stages, when prepared for budding, it clusters toward the cytoplasm of the incipient merozoite [20].…”

“…ELCS can participate in the chromatin autophagy. 16,57,58 A vacuole behind (red arrow) is an autolysosome, lying in the region of centrosome in the emanating microtubules (MT) ( Figure 11A and 11B). Similar to our result, recently, nucleolar aggresomes mediate release of pericentric heterochromatin and nuclear destruction of genotoxically treated cancer cells was reported, and teratocarcinoma PA1 cells treated with etoposide, focusing on the nucleolus most cells display persistent DNA damage and activate macroautophagy.…”
