Differential Roles of C-terminal Eps15 Homology Domain Proteins as Vesiculators and Tubulators of Recycling Endosomes

Abstract: Background: Vesiculation of tubular recycling endosomes is essential for the recycling of receptors and lipids to the plasma membrane. Results: A novel vesiculation assay was used to demonstrate a role for endocytic regulatory proteins in vesiculation. Conclusion: EHD family proteins play significant roles in both the vesiculation and generation of tubular recycling endosomes. Significance: This provides the first direct evidence of differential EHD function in vesiculation and tubulation.

“…(C) local generation of additional membrane-embedded PA further shapes membrane elongation into TRE that contain cargo. D, "Vesiculators," such as EHD1 (33,34), GRAF1 (24) and the lipid modifier cPLA2␣ (33), constrict the TRE by mechanical force and changes in lipid composition, respectively. E, scission gives rise to mobile and small recycling endosomes, which are transported along microtubules toward the PM.…”

“…Nonetheless, we have demonstrated that they have distinct mechanistic functions. EHD1 induces the vesiculation of TRE, whereas EHD3 supports the process of membrane tubulation (11).…”

“…However, a recent study has determined that the GTPase Dyn2 is also required for mitochondrial fission (Lee et al, 2016). Given the homology between EHD1 and dynamin (Daumke et al, 2007), and the role of EHD1 as an ATPase involved in endosomal fission (Cai et al, 2013(Cai et al, , 2014Jakobsson et al, 2011), we sought to explore the possibility that EHD1 may also act directly in mediating mitochondrial fission. To do so, we took advantage of a clever assay used by Lee et al to identify the role of Dyn2 in mitochondrial fission (Lee et al, 2016).…”
