Differential Response of Lung Cancer Cells, with Various Driver Mutations, to Plant Polyphenol Resveratrol and Vitamin D Active Metabolite PRI-2191

Abstract: Plant polyphenols and vitamins D exhibit chemopreventive and therapeutic anticancer effects. We first evaluated the biological effects of the plant polyphenol resveratrol (RESV) and vitamin D active metabolite PRI-2191 on lung cancer cells having different genetic backgrounds. RESV and PRI-2191 showed divergent responses depending on the genetic profile of cells. Antiproliferative activity of PRI-2191 was noticeable in EGFRmut cells, while RESV showed the highest antiproliferative and caspase-3-inducing activi… Show more

“…In our study, we first evaluated the activity of RES against A549 and H1299 cells, which are the most commonly used cell lines in antilung cancer studies (Bian et al, 2022; Fan et al, 2020; J. Li et al, 2022; Y. H. Lin, Zhu, et al, 2021; Maj et al, 2021). Consistent with previous studies (Y. H. Lin, Zhu, et al, 2021), we also demonstrated that RES inhibited the growth and induced apoptosis of NSCLC cells.…”

“…However, many tumors, including lung cancer, can hijack and overactivate the Nrf2/ARE antioxidant system (Sanchez-Ortega et al, 2021;Tian et al, 2016). Importantly, some studies have confirmed that different lung cancer cells respond differently to RES treatment (Maj et al, 2021;Takashina et al, 2017). It is important to understand whether RES inhibits lung cancer by modulating the Nrf2-mediated signaling pathway.…”

“…In lung cancer, PRI-219, a vitamin D3 active metabolite (24R-1,24-dihdroxycholecalciferol), in combination with a hydroxystilbene, RSV (3,5,4′-trihydroxystilbene), induced a significant increase in PD-L1 mRNA expression in NCIH358, A549, and HCC827 cells when compared to PRI-2191 treatment alone [ 50 ]. RSV alone also had a non-significant tendency to upregulate PD-L1 expression in HCC827 cells [ 50 ]. Similarly, RSV was able to increase PD-L1 expression in two lung adenocarcinoma cell lines (A549 and H1299) [ 51 ].…”

“…They also have a low resistance to vitamin D metabolizing enzymes [ 10 ]. The current search is for vitamin D analogs of therapeutic potential against leukemia [ 7 , 11 , 12 ] and solid tumors, including breast [ 13 ], lung [ 14 ], prostate [ 15 ], colorectal [ 16 , 17 ], skin [ 18 ], and ovarian cancer [ 19 ]. We have developed analogs of 1,25D that exceed the potency of the parent 1,25D against cancer cells in vitro and in vivo, and that are less calcaemic and more enzymatically stable [ 10 ].…”
