Differential Response of Hippocampal and Cerebrocortical Autophagy and Ketone Body Metabolism to the Ketogenic Diet

Abstract: Experimental and clinical data support the neuroprotective properties of the ketogenic diet and ketone bodies, but there is still a lot to discover to comprehensively understand the underlying mechanisms. Autophagy is a key mechanism for maintaining cell homeostasis, and therefore its proper function is necessary for preventing accelerated brain aging and neurodegeneration. Due to many potential interconnections, it is possible that the stimulation of autophagy may be one of the mediators of the neuroprotectio… Show more

“…This can prevent neurodegenerative disorders, among other effects, through the elimination of protein aggregates or damaged mitochondria [ 53 , 54 ]. It thus prevents the accumulation of autophagosomes and improves the survival of cortical neurons, leading to a reduction in cerebral damage [ 55 , 56 , 57 ]. Another study has also revealed that ketogenic diets can effectively reduce the amount of beta-amyloid and also other redundant by-products of metabolism in cerebral tissue [ 58 ].…”

“…Additionally, βHB attenuates autophagic degradation and improves neuronal damage induced by N-methyl-D-aspartate (NMDA) in the striatum of rat [ 113 ]. Compared to the cerebral cortex, more autophagosome formation occurs in the hippocampus in response to a KD [ 114 ]. Interestingly, HMGCS2, the control enzyme in ketogenesis, induces autophagic clearance of the β-amyloid precursor protein (APP), which is mediated by acetoacetate metabolism and the mTOR signaling pathway; this suggests that HMGCS2 inhibits the subsequent production and deposition of amyloid β (Aβ) [ 115 ].…”

“…mNGS has both the ability to simultaneously detect DNA and RNA viruses, bacteria, fungi, and parasites present in a sample, and the ability to exclude infections [6]. This technology has gradually been widely used in the eld of pediatric infections, including respiratory system, nervous system, blood system, bone and joint, and parasitic infections [20][21][22][23][24]. Therefore, in this study, mNGS was used to detect the pathogenic bacteria of cholangitis after Kasai operation in biliary atresia, and satisfactory results were obtained.…”
