Transcription promoters of heat shock protein (HSP) genes have been used to control the expression of heterologous proteins in plants and plant cells. To obtain a strong HSP promoter that is functionally active in Nicotiana tabacum BY-2 cells, we set out to identify a promoter of an endogenous gene showing high activation of expression by heat. An N. tabacum BY-2 cell culture was treated for 8 h at 37°C and the cell protein extract analyzed by two-dimensional electrophoresis. A major spot was identified by mass spectrometry as belonging to the small HSP family. The promoter regions and the 5' and 3' untranslated regions of two genes, NtHSP3A and NtHSP3B, with sequences fitting the protein identified were cloned and fused to a hybrid reporter gene coding for β-glucuronidase (GUS) and a yellow fluorescent protein. These constructs were introduced into N. tabacum BY2 cells by Agrobacterium tumefaciens-mediated transformation. Both promoters conferred similar heat-induced GUS expression. In the best heat shock condition, GUS activity was increased 200 fold and reached 285 pmol min(-1) μg protein(-1). Up-scaling in a 4-l bioreactor resulted in similar heat-induced expression. The NtHSP3A promoter was then used to drive the expression of NtPDR1, a plasma membrane transporter belonging to the pleiotropic drug resistance family. No expression was observed at 25°C, while, at 37°C, expression was similar to that obtained using a strong constitutive promoter. These data show that the HSP promoters isolated are useful for high heat-induced expression in N. tabacum BY-2 cells.

Small heat shock proteins (sHSP) are ubiquitously found in all organisms, and with other heat shock proteins (HSP) such as HSP60, HSP70, HSP90, HSP100 made up the molecular chaperone family. They are involved in a wide range of biological processes which include among others cell resistance to biological and environmental stress conditions. In this study, we show by western blotting that CeHSP17, an sHSP of Caenorhabiditis elegans, is significantly induced by high temperatures. Furthermore, in response to metal stress, the CeHSP17 protein expression was significantly induced by cadmium and zinc at high concentration of clearly cytotoxic range in wild-type C. elegans. Altogether, our results show the involvement of CeHSP17 protein in both environmental and biological stresses in C. elegans and establish for the first time the expression pattern of the CeHSP17 protein in response to thermal and metal stress conditions in C. elegans. The responses of CeHSP17 protein expression may serve as potential sensitive biomarker for metal-induced toxicity monitoring and environmental risk assessment.

The 1,053-bp promoter of the oil palm metallothionein gene (so-called MSP1) and its 5' deletions were fused to the GUS reporter gene, and analysed in transiently transformed oil palm tissues. The full length promoter showed sevenfold higher activity in the mesocarp than in leaves and 1.5-fold more activity than the CaMV35S promoter in the mesocarp. The 1,053-bp region containing the 5' untranslated region (UTR) gave the highest activity in the mesocarp, while the 148-bp region was required for minimal promoter activity. Two positive regulatory regions were identified at nucleotides (nt) -953 to -619 and -420 to -256 regions. Fine-tune deletion of the -619 to -420 nt region led to the identification of a 21-bp negative regulatory sequence in the -598 to -577 nt region, which is involved in mesocarp-specific expression. Gel mobility shift assay revealed a strong interaction of the leaf nuclear extract with the 21-bp region. An AGTTAGG core-sequence within this region was identified as a novel negative regulatory element controlling fruit-specificity of the MSP1 promoter. Abscisic acid (ABA) and copper (Cu(2+)) induced the activity of the promoter and its 5' deletions more effectively than methyl jasmonate (MeJa) and ethylene. In the mesocarp, the full length promoter showed stronger inducibility in response to ABA and Cu(2+) than its 5' deletions, while in leaves, the -420 nt fragment was the most inducible by ABA and Cu(2+). These results suggest that the MSP1 promoter and its regulatory regions are potentially useful for engineering fruit-specific and inducible gene expression in oil palm.
