Differential Regulation of LET-7 by LIN28B Isoform–Specific Functions

Abstract: The RNA-binding protein LIN28B plays an important role in development, stem cell biology, and tumorigenesis. LIN28B has two isoforms: the LIN28B-long and -short isoforms. Although studies have revealed the functions of the LIN28B-long isoform in tumorigenesis, the role of the LIN28B-short isoform remains unclear and represents a major gap in the field. The LIN28B-long and -short isoforms are expressed in a subset of human colorectal cancers and adjacent normal colonic mucosa, respectively. To elucidate the fun… Show more

“…Finally, to validate up-regulation of Wnt targets identified via RNA-seq, we used siRNA to knock down IMP1 in Caco2 cells ( Supplementary Fig. 4A,B) that endogenously express LIN28B (Mizuno et al 2018). We observed significant up-regulation of multiple WNT targets with IMP1 knockdown, corroborating our findings in a second, independent cell line (Supplemental Fig.…”

“…However, LIN28B-short isoform lacks the inhibitory ability against the processing by microprocessors. Namely, LIN28B-short isoform functions as a competitor against the LIN28B-long isoform in the context of let-7 maturation [ 25 ].…”

“…Although only a single paper reported that the high let-7g expression was significantly associated with the chemoresistance against S-1 in CRC patients [ 77 ], most of the studies have shown that the downregulation of let-7 contributes to the chemoresistance in the context of CRC. One of the underlying mechanisms by which the downregulated let-7 contributes to the chemoresistance is that the decreased let-7 induces the expression of DNA excision repair protein, excision repair cross-complementing group 1 (ERCC1), which contributes to the resistance against cisplatin or 5-FU [ 25 ].…”

“…This is a random commercial molecule which has been extensively tested in human cell lines by us and others. In addition, it has been widely demonstrated that this control does not alter known functions of miRNAs and mRNAs (54)(55)(56)(57). In this study, the results suggested that this scrambled sequence did not affect the expression of miR-34a or that of EMT-related genes evaluated at 24 and 48 h post-transfection in the MCF-10F and MDA-MB-231 cell lines.…”
