Differential Regulation of Adiponectin Secretion from Cultured Human Omental and Subcutaneous Adipocytes: Effects of Insulin and Rosiglitazone

Abstract: Adiponectin is an adipocyte-derived plasma protein with insulin-sensitizing and antiatherosclerotic properties. Because adipose tissue depots differ in the strength of their association with the adverse metabolic consequences of obesity, we studied the secretion of adiponectin in vitro from paired samples of isolated human omental and sc adipocytes and its regulation by insulin and rosiglitazone. Cells were incubated for 12 or 24 h with and without treatment with 100 nM insulin, 8 micro M rosiglitazone, or bot… Show more

“…21,22 There is also evidence that insulin may have direct effect on adiponectin gene expression and adiponectin concentrations in vitro. [23][24][25] It is therefore conceivable that the higher levels of insulin in insulin-resistant subjects may downregulate levels of adiponectin as suggested previously. 26,27 Although we have shown an inverse association between adiponectin and insulin resistance, it is obvious from Figure 2a that there was overlap of adiponectin levels between insulin-sensitive and insulinresistant patients and some of the insulin-sensitive individuals had low adiponectin.…”

“…Adiponectin is an adipokine which decreases when fat accumulates [20]. In this paper, adiponectin was observed as an independent protective factor against MetS and its associated components, owing to the inverse relationship of adiponectin with triglycerides and fasting glucose, and the positive association of adiponectin with HDL cholesterol.…”

“…Smokeless tobacco and alcohol consumption have been shown to have a role in obesity and cardiometabolic diseases [18,19]. With increasing fat accumulation, the level of adiponectin (a hormone secreted from adipose tissue) is reduced, suggesting that it may have a role in MetS [20]. Identifying the risk factors influencing body composition and MetS is fundamental to understanding the pathogenesis of these diseases in this group of African women, who seem to be more susceptible to this phenomenon than white women [7].…”

“…Differences statistically significant compared to non-pregnant with p<0.001 ISI Insulin sensitivity index, HGP hepatic glucose production a Pregnant vs pregravid tion of plasma adiponectin. Total adiponectin, as well as HMW multimers, decrease in response to insulin infusion [6,35,36], but this may be regulated at the level of secretion rather than transcription [37]. Adiponectin is secreted as HMW and LMW multimeric complexes, and the amount of circulating complexes is likely to be regulated at the tissue level because LMW and HMW complexes do not interchange in the serum [38].…”
