Differential Regulation in the Expression of Hepatic Genes in Nephrotic and Pair-Fed Rats

Abstract: The messenger ribonucleic acid (mRNA) levels of albumin, fibrinogen, α1-acid glycoprotein (pAGP) and transferrin were analyzed in puromycin aminonucleoside (PAN)-treated (nephrotic) and in pair-fed (PF) rats with the Northern and dot blot hybridization techniques. Albumin mRNA levels in nephrotic and PF rats were 2- and 1.5-fold higher, respectively, than in ad-libitum-fed control (C) rats 6 days after PAN treatment. On day 11, this mRNA in PAN-treated rats was 2.5-fold higher than in PF and 4-fold higher than… Show more

“…No changes were found in the mRNA level or transcription rate for this protein. Results similar to these have also been found in PAN nephrosis (23). Therefore, nephrosis does not increase the hepatic synthesis of all secretory proteins to the same extent, and the acute phase response, if involved at all, is not operative after the establishment of proteinuria.…”

“…Tile ACE activity lneasurements for testis and epididymis were performed using the linear segments of the plots of HA production vs tissue extract volume and incubation time. The ACE activities of the testis and epididymis were inhibited by HHL at concentrations above 5 mM just as is the ACE of human heart muscle (15), testis (7) and sheep serum (16). Eventhough the K values and HA concentrations obtained were different for each tissue, the inhibition by HHL occurred at concentrations above 5 raM.…”

“…Although protein C and AT have similar small molecular size, protein C has a negative charge that markedly reduces its relative renal loss. More importantly, increased hepatic synthesis of protein C and other vitamin K ‐dependent zymogens has been documented in a rat model of nephrotic syndrome 63 . Protein C shares common transcription regulatory sites with the other vitamin K ‐dependent coagulation proteins, including at least 3 liver‐specific regulatory elements 64 .…”
