Differential proteomic analysis of developmental stages of Acca sellowiana somatic embryos

Abstract: Feijoa (Acca sellowiana, Myrtaceae), a native fruit species from southern Brazil and northern Uruguay, is considered to constitute a reference system for somatic embryogenesis in woody dicots. This in vitro regenerative pathway is an efficient micropropagation method, and a suitable model system for studies in plant developmental physiology. This study attempts to detect and identify proteins that are expressed during the different developmental stages of somatic embryos of A. sellowiana. Using high resolution… Show more

“…The total proteins levels of somatic embryos increased according to the progression in the developmental stages (Figure 3 B), confirming the same pattern of behavior for the accumulation of proteins in this same species (Cangahuala-Inocente et al, 2009). The protein levels ranged from 0.1 mg g -1 FW in the heart stage to 0.7 mg g -1 FW in the cotyledonary stage.…”

“…As proteins are involved in regulating cell expansion and creation of biophysical characteristics required for seedling morphogenesis (Jiménez 2001), they are commonly consumed during induction of embryogenesis being available for the redifferentiation of somatic cells (Cangahuala-Inocente et al 2009). The lipids, together with proteins, play a key role in the composition of cell membranes (Kraus et al 2012).…”

“…The histochemical methods allow the analysis of the mobilization and synthesis of reserves during embryogenic development, allowing recognition of regions of high energy demand (Branca et al 1994;Cangahuala-Inocente et al 2009;Rocha et al 2012).…”

“…Furthermore, there has been recent research on the proteome of embryonic callus and non-embryogenic callus of Vitis vinifera (Zhang et al, 2009;Marsoni et al, 2008). A high resolution 2-DE proteome reference map was generated for different developmental stages of Acca sellowiana somatic embryos (Cangahuala-Inocente et al, 2009). In Cyclamen persicum, Winkelmann et al (2006) detected 200 protein spots in zygotic and somatic embryos using high resolution 2-DE and sodium dodecylsulfate polyacrylamide gel electrophoresis (SDS-PAGE) (Winkelmann et al, 2006).…”
