Differential photodynamic response of cultured cells to methylene blue and toluidine blue: role of dark redox processes

Abstract: Cultured cells treated with equal concentrations of thiazine photosensitizers methylene blue (MB) or toluidine blue (TB) showed a distinct photodynamic lethality, with TB being much more effective, when exposed to red light from a LED source. This effect is accounted for because of the differences in the chemical reduction of MB and TB in the intracellular environment. While TB accumulates as blue granular structures, MB does not give such a localization pattern. However, upon exposure of MB-treated cells to o… Show more

“…The contact of MB with biological environments containing reductase enzymes usually causes a small but reversible chemical reduction of the dye leading to an increase in the concentration of leucoMB 17,18. In our experimental conditions, this effect was visualized by putting MB in contact with E. coli and observing an increased absorption at 290 nm and a decrease in the absorption peak at 660 nm.…”

“…[10] TB has demonstrated a strong singlet oxygen quantum yield and a favored absorption (633 nm) peaked in the far-red window (600–900 nm). [11] The ideal cationic-aromatic structure of TB makes it able to be noncovalently packaged into HA to form a HA/TB nanogel using a recently developed protocol. [8c] The quenched TB fluorescence in HA/TB aids in the convenient to monitoring of the intracellular release of TB.…”
