Differential Gene Expression in the Bovine Corpus Luteum During Transition from Early Phase to Midphase and Its Potential Role in Acquisition of Luteolytic Sensitivity to Prostaglandin F2 Alpha1

Abstract: Prostaglandin F2 alpha (PGF(2alpha)) brings about regression of the bovine corpus luteum (CL). This luteolytic property of PGF(2alpha) is used in beef and dairy cattle to synchronize estrus. A limitation of this protocol is insensitivity of the early CL to luteolytic actions of PGF(2alpha). The mechanisms underlying this differential luteal sensitivity are poorly understood. The developing CL has a maximum number of PGF(2alpha) receptors; therefore, differences in signaling events may be responsible for luteal… Show more

“…The use of GAPDH as a housekeeping gene has been previously validated [36], and we further validated the consistency of steady-state concentrations for this gene in the CL in preliminary experiments (unpublished results). This preliminary experiment indicated similar expression of GAPDH after multiple biopsies of the same animal and similar concentrations in the different treatment groups and different collection times in the present experiment.…”

“…Sequences for primers were designed using Primer3 from the NCBI (http:// www.ncbi.nlm.nih.gov/) gene database or were derived from published studies [16,22,[35][36][37]. Primer pair sequences and product sizes are shown in supplemental Table S1 (available online at www.biolreprod.org).…”

“…EGR-1 and c-Fos have been shown as important key players of luteolytic acquisition in pig and cows (Chen et al 2001, Hou et al 2008, Atli et al 2012, Diaz et al 2013. PGF 2a -FP interaction increases intracellular Ca 2C which activates multiple cell signaling pathways and mediates the acquisition of luteolytic sensitivity to PGF 2a in the bovine luteal cells (Goravanahally et al 2009, Wright et al 2014. Interactions among PGF 2a , Ca 2C , PKC, ERK1/2, c-Fos, and c-JUN have been shown in luteal cells in cows (Chen et al 2001, Yadav et al 2005.…”

“…Tissues from Day 4 (n ÂĽ 4) and Day 10 (n ÂĽ 4) bovine CL were homogenized individually and total RNA was isolated using Tri-reagent according to the manufacturer's instructions (Molecular Research Center) and as previously described [36]. Briefly, frozen CL tissue immersed in liquid nitrogen was pulverized mechanically using an RNase-free mortar and pestle.…”
