Differential expression of up-regulated cofilin-1 and down-regulated cofilin-2 characteristic of pancreatic cancer tissues

Abstract: Abstract. Pancreatic cancer (PC) is one of the most deadly malignant tumors. The aim of this study was to identify potential biomarkers for PC. Using two-dimensional gel electrophoresis and liquid chromatography-tandem mass spectrometry, the proteomic profiles of pancreatic cancerous and non-cancerous tissues from ten patients with PC were compared. One of the numerous spots that showed stronger intensity in cancerous compared to non-cancerous tissues was identified as non-muscle cofilin (cofilin-1). This up-r… Show more

“…It has also been reported that more than one form of cofilin exist in pancreatic cancer tissues [31]. Cofilin-1 involved in motility and invasion is up-regulated in pancreatic carcinoma tissues while muscle cofilin-2 was reported to be downregulated [31]. At present, it is not clear whether O-DDHSL directly interact with these molecules, however, its effect seem to vary between the cells tested in this study.…”

“…RhoC is a small GTPase which is an important effector of tumor cell motility [28] and is expressed in pancreatic tumors [30]. Similarly, cofilin is also present in pancreatic carcinoma tissues and possibly promotes its progression [31].…”

“…Cofilin-1, a small protein able to bind actin, has been previously shown to be overexpressed in some tumors [40][42]. In oral carcinomas, few studies have observed altered expression of cofilin [32][34] but no reference about its role on oral cancer cell invasion has been published.…”

“…The three isoforms of mammalian cofilins are indispensable and vary in distribution in normal and diseased tissues32728, abilities to support cellular activities2930, and in their affinities for G- and F-actin393132. Among these, only CFL2, the muscle-specific isoform of ADF/cofilins, was characterized in complex with F-actin by cryo-EM reconstruction at 9 Å resolution13, and therefore, was selected for this study.…”
