Differential expression of the psbB and psbH genes encoding the 47 kDa chlorophyll a-protein and the 10 kDa phosphoprotein of photosystem II during chloroplast development in wheat

Abstract: The nucleotide sequence of a region of wheat chloroplast DNA containing the psbB gene for the 47 kDa chlorophyll a-binding protein of photosystem II has been determined. The gene encodes a polypeptide of 508 amino acid residues which is predicted to contain six hydrophobic membrane-spanning regions. The psbB gene is located 562 bp upstream of the psbH gene for the 10 kDa phosphoprotein of photosystem II. A small open reading frame of 38 codons is located between psbB and psbH, and on the opposite strand the ps… Show more

“…On one hand, the Chl a/b-proteins of PSII show a lower ratio of Chl to protein and a higher Chl-a/b ratio. As repeatedly shown, Chl is necessary for the stabilization of most Chl-protein complexes (Mathis and Burkey 1989;Henry et al 1990;Hoober et al 1990;Mullet et al 1990;Hird et al 1991). However, as our data indicate, the full setup is not required.…”

“…Chlorophyll, however, seems to be necessary for stabilizing a number of Chl-binding apoproteins of the PSII core-complex (Mullet etal. 1990;Hird et al 1991) as well as the light-harvesting complex II (LHCII; Mathis and Burkey 1989;Henry et al 1990;Hoober et al 1990). These proteins are not found in dark-grown plants and in plants from IL the size of the Chl-antenna is reduced (Argyroudi-Akoyunoglou and Akoyunoglou 1979; Dubertret and LefortTran 1981;Day et al 1984;White and Green 1988;Morrissey et al 1989).…”

“…Their total lengths were 14.0 and 13.7 kb, respectively. The ctDNA regions included the 3Ј flanking region of the Rubisco large subunit gene (rbcL) (10) and 10 structural genes: encod-ing ATP synthase, ␣, ␤, and subunits (atpA, atpB, and atpE) (11), ATP synthase, CF 0 subunit III and CF 0 subunit I (atpH and atpF) (12), the 47-kDa chlorophyll a-binding protein, the 10-kDa phosphoprotein of photosystem II, photosystem II polypeptide of 43 aa, apocytochrome b-563, and the 15-kDa polypeptide of the cytochrome b-f complex (psbB, psbN, psbH, petB, and petD) (13). The mtDNA regions include nine structural genes: encoding the cytochrome oxidase subunit I, II, and III (cox1, cox2, and cox3) (14-16), the ATP synthase ␣ subunit (atpA) (17), NADH dehydrogenase subunit I (nad1-a, nad1-b, and nad1-e) (18), cytochrome b (cob) (19), and ORF25 (orf25) (20).…”

“…The PsbH polypeptide was present in both etiolated tissues and deetiolated leaves, whereas the PsbB subunit was absent from dark-grown wheat plants but became detectable on greening. Phosphorylation of the PsbH polypeptide occurred even in etioplasts (Hird et al 1991).…”
