Differential expression of HLA‐DR and DR‐linked determinants on human leukemias and lymphoid cells

Abstract: Leukemic and normal hemopoietic cells were examined with the monoclonal anti-bodies DA2 and Genox 353 for the presence of HLA-DR and DR-linked (DC/MB) determinants, respectively. Although most non-T acute leukemias and leukemic cell lines expressed the monomorphic DR determinant detected by DA2, fewer than expected expressed the DR-linked polymorphic specificity detected by Genox 353. TdT+ lymphoid precursors from normal bone marrow were also DA2+ but Genox 353-. T cells and thymocytes which were DA2-, Genox 3… Show more

“…It is of interest to note that the DC/DS determinant detected by Genox 3.53 is not expressed on AML blasts and precursor cells of the erythroid and myeloid series (4,5). In addition, Genox 3.53 fails to stain TdT + bone marrow cells and only 20% of the pre-B cells are positive (6). It appears that DC/ DS expression is limited to more mature stages of B cell differentiation.…”

“…The recently described HLA-linked DC/DS antigens (2, 3) have more limited cell distribution (4)(5)(6) and are expressed predominantly on B cells, activated T cells, and a minor population ofmonocytes. Although their expression is documented in cases of B cell chronic lymphocytic leukemia (CLL), they are infrequently found on cells of acute lymphoblastic leukemia (ALL) of non-T cell type (4).…”

“…Non-coordinate expression of DR and DQ antigens is also a feature of a number of established lines derived from cases of ALL Newman et al, 1983;Wang et al, 1983). That is, the majority of cells from the lines are DRpositive and fewer cells are DQ-positive.…”
