Abstract-Induction of the cytochrome P4501A (CYP1A) enzyme system in fish is a common biomarker of exposure to aromatic hydrocarbons. Induction of CYP1A can be measured at a number of steps in the transcription-translation-functional protein pathway using a variety of techniques. The present study examined the range of these measurements from 94 published papers in an attempt to examine the statistical characteristics of each method. Cytochrome P4501A induction, as measured by catalytic ethoxyresorufin-O-deethylase (EROD) activity, protein levels (enzyme-linked immunosorbent assay, Western blot analysis, and immunohistochemistry), and mRNA levels (Northern blot analysis and reverse transcription-polymerase chain reaction), was analyzed. When possible, the variance structure, effect size determination, and dose-response modeling of each method of measurement in the laboratory and field were examined. Conclusions from this analysis include: 1) Because of interlaboratory and interspecies variability, general end-point determinations will need to be defined in terms of the statistically detectable fold-change of measurements relative to control or reference values, and 2) fold-change in EROD activity provides the most robust measure of the dose responsiveness of aromatic hydrocarbons within specific chemical classes (e.g., polycyclic aromatic hydrocarbons). The relationship between the ability to measure statistical differences in induction level and the biological significance of those measurements has yet to be defined. To utilize these biomarkers in a risk assessment context, this relationship must be addressed at the scientific and management levels.

As long-lived marine mammals found throughout the temperate coastal waters of the North Pacific and Atlantic Oceans, harbour seals (Phoca vitulina) have become an invaluable sentinel of food-web contamination. Their relatively high trophic position predisposes harbour seals to the accumulation of harmful levels of persistent organic pollutants (POPs). We obtained skin/blubber biopsy samples from live-captured young harbour seals from various sites in the northeastern Pacific (British Columbia, Canada, and Washington State, USA) as well as the northwestern Atlantic (Newfoundland and Quebec, Canada). We developed harbour seal-specific primers to investigate the potential impact of POP exposure on the expression of eight important genes. We found correlations between the blubber mRNA levels of three of our eight target genes and the dominant persistent organic pollutant in seals [polychlorinated biphenyls (PCBs)] including estrogen receptor alpha (Esr1: r = 0.12, p = 0.038), thyroid hormone receptor alpha (Thra: r = 0.16; p = 0.028), and glucocorticoid receptor (Nr3c1: r = 0.12; p = 0.049). Age, sex, weight, and length were not confounding factors on the expression of genes. Although the population-level consequences are unclear, our results suggest that PCBs are associated with alterations of the expression of genes responsible for aspects of metabolism, growth and development, and immune function. Collectively, these results provide additional support for the use of harbour seals as indicators of coastal food-web contamination.

Alterations in histopathology and stress-associated gene expression induced by infection with Prohemistomum vivax encysted metacercariae in Nile tilapia

Due to limited data on the pathogenicity of Prohemistomum vivax (P. vivax) and its impacts on fish health, this study aimed to determine the morphological, molecular characteristics, pathogenicity, and histopathological alterations in fish infected with P. vivax. Eight hundred (800) Nile tilapia (Oreochromis niloticus) were collected from various farms in Kafr El Sheikh Governorate. The fish were examined for encysted metacercariae (EMC) in different organs. Tissue specimens were collected and underwent histopathological analysis, expression of stress-related genes, and genetic characterization by sequencing of the internal transcribed spacer 2 (ITS2). P. vivax metacercariae were oval to round in shape and were collected from various organs including the muscle, skin, eyes, intestine, liver, kidney, and gills of infected O. niloticus. Sequencing and phylogenetic analysis of the ITS2 region revealed a 507-bp fragment, confirming parasite identity and matching within the same clade as other P. vivax isolates. Infected fish displayed abdominal hydropsy, skin darkening, and emaciation. P. vivax encysted metacercariae were detected during the study period in 620/800 fish, with an overall prevalence of 77.5%. The seasonal prevalence was 95% in summer, 85% in spring, 55% in autumn, and 75% in winter. The intensity of infection was 1–40 cysts per microscopic field. Histopathological examination of muscles revealed parasitic cysts embedded within muscle fibers, causing severe degeneration and necrosis. Upregulation of cytochrome P450 (cpy1a1), heat shock protein 70 (hsp-70), and tumor suppressor p53 (p53) was recorded in both liver and muscle samples of infected tilapia compared to controls. This indicates activation of detoxification, cellular stress, and apoptotic pathways in response to P. vivax infection. There is limited data available on the pathogenicity of P. vivax and its impacts on fish health; thus, this study provides key insights into the morphology, pathogenicity, and histopathological impacts of P. vivax in Nile tilapia.
