Differential expression of ABH histo-blood group antigens and LAMPs in infantile hemangioma

Abstract: Although infantile hemangioma (IH) are the most common tumors of infancy, the mechanism of their proliferation and involution remains vague. Proliferation, differentiation and death of endothelial cells are the basic processes involved in their pathobiology. Here we hypothesize that the glycoconjugates ABH histo-blood group antigens (HBGA) and lysosome-associated membrane proteins (LAMPs) might be implied in both the differentiation and death of endothelial cells during vascular remodeling in IH. Proliferating… Show more

“…Furthermore, lysosome-associated membrane proteins (LAMPs), proteins associated with autolysosome formation, were found in immature endothelium of proliferating infantile hemangioma, a tumor of the microvasculature characterized by aggressive angiogenesis during infancy. LAMPs were increased in involuting hemangioma, which might be associated with augmented autophagy required for tissue remodeling during tumor involution (36). Additionally, cathepsin D, a lysosomal hydrolase associated with autophagy, has been found to enhance angiogenesis of cancer cells probably by degrading extracellular matrix (20).…”

“…It was shown that a histo-blood group B-like antigen is strongly upregulated during rat mammary gland involution, implying that this antigen might either provide a survival function and/ or be expressed in epithelial cells destined to remodel mammary ducts (Mengwasser and Sleeman 2001). These molecules are also implicated in the death of endothelial cells in regressing infantile hemangioma (Sarafian et al 2005). The current study indicates that HGBAs are expressed not only in human and animal tissues undergoing cell death but also in cell cultures exposed to toxic agents.…”

“…The indirect immunoperoxidase technique using the universal LSAB-2 kit (Dako, Glostrup, Denmark; cat no. K0672) with chromogen aminoethylcarbazole (AEC) was carried out according to previously described protocols (Sarafian et al 2005). Cell proliferation was assessed using monoclonal antibodies with defined specificity to human antigens: PCNA (proliferative cell nuclear antigen) (Dako; cat no.…”

“…As endothelial cells are continuously hit by circulating cells, they are believed to be among the cells most frequently wounded and repaired (1). Our previous investigations suggest that ABH histo‐blood group antigens (HBGAs) might be used as markers for endothelial differentiation of embryonic mesenchymal cells (2), as well as indicators of endothelial cells in vascular tumors such as infantile hemangioma (3). As there are few investigations of these molecules in the granulation tissue it seemed interesting to follow their expression in sprouting capillaries.…”
