Differential expression changes in K562 cells during the hemin-induced erythroid differentiation and the phorbol myristate acetate (PMA)-induced megakaryocytic differentiation

Abstract: K562 cell line has been used as a model of common progenitor of erythroblasts and magakaryocytes and can be differentiated into erythroid and megakaryocytic lineages by hemin and phorbol myristate acetate (PMA) respectively. We analyzed mRNA expression in un-induced, hemin-induced and PMA-induced K562 cells by differential display reverse transcription polymerase chain reaction (DDRT-PCR) method. 314 differential expression sequence tags (ESTs) were obtained. Among them, 201 ESTs displayed up-regulation and 85… Show more

“…Therefore, differentiation of K562 cells has served as a representative model of studies of erythroid differentiation for many years (Huo et al, 2006;Wang et al, 2009). The undetectable expression of AE1 in K562 cells suggests that miR-24 may take part in the tumorigenesis of erythroleukemia by arresting erythroid maturation through silencing of AE1 expression.…”

“…These processes are associated with a series of molecular events. The molecular events that govern these processes are only partially understood [34]. The function of erythrocyte is oxygen delivery.…”

“…Obtaining synchronized cells from long-term cultured proerythroblast cells is very difficult which led us to use K562 cells, which were suitable for this study. The K562 cell line has been used as a model of common progenitor of erythroblasts and megakaryocytes and can be differentiated into erythroid and megakaryocytic lineages by hemin (ferriprotoporphyrin IX) and phorbol myristate acetate (PMA) respectively [34].…”

“…Hemin chloride 40 uM (H-5533 Sigma; St Louis, MO, USA) and theophylline 7 mM (T-1633 Sigma; St Louis, MO, USA) were used as positive differentiation inducers for K-562 (Huo et al, 2006) and Daudi (Sandlund et al, 1993) cell lines, respectively. Triplicates of cell suspension of each cell line (10 6 cell/mL) in complete RMPI 1640 medium containing ARF solution to give final concentrations ranging from 0.1 to 100 µg/mL, or the required concentration of the cell differentiation inducers, in a 1000 µL final volume/well were distributed in 24-well culture plates (TPP; Switzerland) and incubated for 7 days at 37 °C in 5 % CO 2 .…”
