Differential EEG effects of the anxiolytic drugs, deramciclane (EGIS-3886), ritanserin and chlordiazepoxide in rats

Abstract: The influence of serotonergic and benzodiazepine type anxiolytic drugs on the cortical activation and sleep-wakefulness cycle were compared by evaluating the effects of ritanserin and deramciclane (EGIS-3886), two 5-HT2 receptor antagonists, and chlordiazepoxide on the electroencephalogram (EEG) in freely moving rats. Following drug administration (1, 3, and 10 mg/kg, PO for all drugs), EEG was continuously sampled for 6 h and power spectra were calculated for every 5 s to assess changes in slow wave activity … Show more

“…These objective thresholds were then used to score recordings obtained after the treatments. Epochs in which delta power was above and EMG value below these thresholds, respectively, were marked as SWS, while epochs with lower delta power or higher EMG activity as W. Scoring was performed by a semi-automated method had been published earlier 84 and was used in several studies published by our laboratory later 50,68,69,[85][86][87] . Raw hypnograms were smoothed, i.e.…”

“…However, some drugs, e.g. benzodiazepines, selectively decrease delta power without interrupting sleep, causing a shift from deep sleep to light sleep (Détári et al, 1999). Bensultap acts on nicotinic acetylcholine receptors, not on GABA receptors as do benzodiazepines, but a similar effect on delta power cannot be excluded.…”
