Differential disposition of intra-renal generated and preformed glucuronides: studies with 4-methylumbelliferone and 4-methylumbelliferyl glucuronide in the filtering and nonfiltering isolated perfused rat kidney

Abstract: The data confirmed an important role for the kidney in the metabolic clearance of xenobiotics via glucuronidation and signalled the lack of impact of impaired glomerular filtration on renal drug metabolism.

“…Moreover, it is conceivable that CNF may inhibit renal UGT1A9 activity in vivo to a greater extent than predicted from the [I]/K i,u ratio. We have demonstrated that basolateral uptake of 4MU in the isolated rat perfused kidney is high, resulting in extensive renal 4MU glucuronidation (Wang et al, 2011). Thus, the intrarenal concentration of drugs, such as CNF (and DPF), may be higher than in plasma, resulting in a larger than expected [I]/K i,u ratio.…”

“…The curcumin metabolites that have been basolaterally exported from enterocytes, hepatocytes, or nonenterohepatic organ cells are most likely to undergo renal clearance, given that the kidneys are responsible for the elimination of Cancer Pharmacology of Curcumin a myriad of conjugated compounds, including xenobiotic metabolites (Lohr et al, 1998). Although renal uptake and clearance of curcumin conjugates has never been directly demonstrated, it does constitute the most favorable route inasmuch as renal uptake increases with a compound's hydrophilicity, charge, and Hbonding capacity and kidney cells are equipped with a plethora of basolateral importers and apical exporters (Kusuhara and Sugiyama, 2009;Morrissey et al, 2013) to eliminate the conjugated compounds via urine (Wang et al, 2011a).…”

“…Vanillin, which is a food-flavoring agent utilized widely in foods, beverages, drugs, and perfumes, was also found as one of the main oxidation compounds in oil palm EFB lignin. [404142434445464748495051525354] Previous study has investigated the UGT conjugation pathway of vanillin by liver microsomes from human, dog, rat, mice, minipig, and cynomolgus monkey. [55] The study showed that vanillin was efficiently glucuronidated by liver microsomes obtained from various species.…”
