Differential configurations involving binding of USF transcription factors and Twist1 regulateAlx3promoter activity in mesenchymal and pancreatic cells

Abstract: During embryonic development, the aristaless-type homeodomain protein Alx3 is expressed in the forehead mesenchyme and contributes to the regulation of craniofacial development. In the adult, Alx3 is expressed in pancreatic islets where it participates in the control of glucose homoeostasis. In the present study, we investigated the transcriptional regulation of Alx3 gene expression in these two cell types. We found that the Alx3 promoter contains two E-box regulatory elements, named EB1 and EB2, that provide … Show more

“…Temporal changes in USF2 and TWIST2 transcript abundance were established and compared to results obtained for USF1 to determine potential association of these transcription factors (Garcia-Sanz et al 2013) in transcriptional regulation of JY-1 , GDF9 and other genes containing multiple E-box binding elements in their promoter regions. In comparison with USF1 , USF2 transcript abundance displayed similar changes during oocyte and early embryonic development (Fig.…”

“…Most commonly, this transcription factor binds to a pyrimidine-rich initiator (Inr) elements or E-box motifs of the promoter region as homodimers or a heterodimer (USF1/2) and might act with or without association with other E-box binding transcription factors of bHLH family (Garcia-Sanz et al 2013). The USF1/USF2 heterodimer is a major DNA binding form present in most tissues and cell types compared to the homodimers (Fair et al 1995, Rajput et al 2013).…”

“…Other mechanisms have implications with regard to cell growth control. USF1/2 interact with components of the basal transcription complex [Garcia-Sanz et al, 2013] and guide recruitment of the hSET1A histone methylase complex to chromatin as part of lineage-specifc cell differentiation [Deng et al, 2013]. Recent findings suggest, moreover, that USF2 is required for induction of HIF2α responsive genes (including PAI-1) under hypoxic conditions likely involving recruitment of the histone acetylases p300 and CBP [Palmeri et al, 2002].…”
