Objective: The objective is to observe the treatment effect of electro-acupuncture (EA) on core circadian clock gene Per2 and Bmal1 expression in hypothalamus of sleep-deprivation (SD) rats. Methods: Thirty-two Wistar male rats were randomly divided into 4 groups. Mice in the blank control group did not receive any treatment; the remaining groups were applied with para-chlorophenylalanine (PCPA) 300 mg/kg intraperitoneal injection for 2 days. Diazepam group received intraperitoneal injection of Diazepam (0.9 mg/kg, i.p.) one time a day for 5 days, while M group was treated with saline (0.9 mg/kg, i.p.) at the same time. Rats in EA group were given EA treatment, 20 minutes, once a day for 5 days, and rats in remaining groups were put into fixation-machine for the same time everyday, lasting for 5 days. Rats were sacrificed after anesthesia at the 8th day. Real-time PCR was adopted to detect the expression in clock gene Per2 and Bmal1 of each group. Results: Compared with blank control group, the expression of Per2 was significant decreased in PCPA model group (P < 0.05), the expression of Bmal1 was increased in PCPA model group (P < 0.05). Compared with PCPA model group, the expression of Per2 were significant enhanced in EA group and Diazepam group (P < 0.05). Simultaneously, compared with PCPA group, the expression of Bmal1 was no statistically significant in EA group and Diazepam groups (P > 0.05). Conclusion: EA can significant up-regulate the expression of Per2 in SD rats, and down-regulate gene Bmal1 expression, and benefiting the weight of rats. Thus, EA is a potentially promising intervention to treat sleep-deprivation.

This study aims to compare the effectiveness of acupuncture and medication therapy on improving sleep disorders in women suffering from Generalized Anxiety Disorder. This study is a quasi-experimental with pretestposttest, control group, and follow-up period. The study population included all women suffering from generalized anxiety disorder and sleep disorder. Purposive sampling was used to select the sample. Using this method, 45 participants were selected and randomly assigned to three groups: acupuncture, medication therapy, and control groups. Acupuncture group received 12 to 15 treatment sessions (2-3 times a week) in DU20, HT7, PC6, Kid6, Ren12, UB62, Sp6, Anmian and Ear-shen men. Medication therapy group were taken drugs under the supervision of a psychiatric while the control group received no intervention over this period. In order to analyze the data, repeated measures were used in the statistical SPSS software version 21. The results showed that both acupuncture and medication therapy programs improve sleep disorder in women with generalized anxiety disorder; however, the acupuncture was more effective than medication therapy. Background:Traumatic brain injury (TBI) has become a major cause of morbidity and mortality both in China and abroad. Disorders of consciousness (DOC) following severe traumatic brain injury (TBI) is a common refractory complication, resulting in difficult rehabilitation and poor life quality. However, effective therapeutic approaches remain limited. Although auricular-acupuncture has been widely applied in the treatment of neurological disorders in China, its efficacy and safety for consciousness recovery remain to be elucidated. Methods: Here, we conduct a study design and protocol of a randomized, blinded, controlled study to evaluate the efficacy and safety of Electroacupuncture at Auricular Acupoints “Heart” and “Brainstem” in the consciousness recovery of patients with TBI. A total of 80 patients with initial Glasgow coma scale score between 3-8 points will be recruited in the trial and randomized into treatment (combined application of auricular and body acupuncture) group or control (body acupuncture alone) group. Patients in the treatment group will receive electroacupuncture at bilateral auricular acupoints “Heart” and “Brainstem” (four points in total) combined with body acupuncture in a supine position while patients in the control group will receive body acupuncture alone for 4 weeks. The primary outcomes are changes of Glasgow coma scale score and mismatch negativity of event-related brain potentials before and after treatment. The secondary outcome measures will be changes of Barthel and FuglMeyer scores. The safety will also be assessed by monitoring the incidence of adverse events and changes in vital signs during the study. Discussion: Results from this trial will significantly support the application of auricular acupuncture in the consciousness recovery of patients with severe TBI. If found to be effective and safe, auricular acupuncture will be a valuable complementary option for comatose patients with TBI. Trial registration: Chinese Clinical Trial Registry: ChiCTR1800020245.
