Different effect of resveratrol to induction of apoptosis depending on the type of human cancer cells

Abstract: The effect of resveratrol on various human cancer cells was investigated with special focus on apoptotic cell death, in an attempt to further characterize its mechanism of action. There were great differences in the anti-viability effect of resveratrol between different types of human cancer cells. While the inhibition of cell viability by resveratrol was marked in U937 and MOLT-4 leukemia cells, resveratrol moderately inhibited cell viability in MCF-7 breast, HepG2 liver, and A549 lung cancer cells, and the e… Show more

“…After analyzing these five pathways in CMap, several known compounds were identified. Unsurprisingly, many among the top 10 have been studied for the treatment of LUAD, including etoposide, irinotecan, resveratrol, and daunorubicin . This finding confirms that several well‐known anti‐cancer drugs used in LUAD treatment fulfill their function by targeting the five aforementioned signaling pathways.…”

“…The cytotoxicity test did not reveal harmful effects due to RSV or tBHQ and based on the results of the cytotoxicity assay, morphological analysis (data not shown) and data in the literature (Gharavi & El‐Kadi, ; Li et al, ; Plauth et al, ; Sticozzi et al, ; Takashina et al, ), the further experiments have been performed at the dose of 10 µM for both compounds.…”

“…RSV was dissolved in absolute EtOH to obtain a 10 mM stock‐solution and tBHQ was prepared in DMSO to obtain a 100 mM stock solution. Based on the cytotoxicity experiments and the literature (Gharavi & El‐Kadi, ; Li et al, ; Plauth et al, ; Sticozzi, Cervellati, Muresan, Cervellati, & Valacchi, ; Takashina et al, ), the experiments were performed at 10 µM for both compounds.…”

“…Percentage of necrotic cell significantly increased at 5 and 10 mM concentrations when compared to control group. Early stage apoptosis is represented by disruption of mitochondrial membrane potential, but cells maintain membrane integrity [29]. Since mitochondrial dysfunction is an early event in the process of apoptosis, early apoptotic cells arise from a reduced mitochondrial transmembrane potential that may precede the release of cytochrome c from the mitochondria.…”

“…Since mitochondrial dysfunction is an early event in the process of apoptosis, early apoptotic cells arise from a reduced mitochondrial transmembrane potential that may precede the release of cytochrome c from the mitochondria. On the other hand, the late stages of apoptosis are characterized by DNA fragmentation and loss of cell membrane permeability [29]. Thus, a feature of late apoptosis is large fragmentation of genomic DNA that produces multiple DNA doublestrand breaks with accessible 3′-hydroxyl groups.…”
