Differences in Potency of CXC Chemokine Ligand 8-, CC Chemokine Ligand 11-, and C5a-Induced Modulation of Integrin Function on Human Eosinophils

Abstract: The hypothesis was tested that different chemoattractants have different effects on the activity of integrins expressed by the human eosinophil. Three chemoattractants, CXCL8 (IL-8), CCL11 (eotaxin-1), and C5a were tested with respect to their ability to induce migration and the transition of eosinophils from a rolling interaction to a firm arrest on activated endothelial cells under flow conditions. CCL11 and C5a induced a firm arrest of eosinophils rolling on an endothelial surface, whereas CXCL8 induced onl… Show more

“…11). ␣ M ␤ 2 is believed to be important for eosinophil migration (22,51,53,(93)(94)(95). Thus, the scenario has migrating eosinophils using activated ␣ M ␤ 2 and possibly other integrins to interact with multiple substrates (22,36,92), including VCAM-1, ICAM-1, and extracellular matrix proteins, on endothelial cells, in connective tissue, and on epithelium of the bronchial wall.…”

“…Because the GTPase Rap1 has been shown to play a role in several cases of agonist-induced integrin activation (4,20,21,36), we tested the hypothesis that Rap1 may also regulate basal ␣ 4 -integrin function in resting eosinophils. Active GTP-bound Rap1 was readily precipitated from lysates of resting eosinophils (Fig.…”

“…Mononuclear cells were removed by centrifugation over isotonic Ficoll (1.077 g/ml). After lysis of erythrocytes with an isotonic icecold NH 4 Cl solution, granulocytes were washed and resuspended in isolation buffer. Eosinophils were purified from granulocytes by negative immunomagnetic selection using anti-CD16-conjugated microbeads (MACS; Miltenyi Biotec) (23) and anti-CD3 and -CD14-conjugated microbeads were additionally added to avoid mononuclear cell contamination.…”

“…Since C5a can affect regulation of the ROCK pathway, surface expression of integrin and cadherin family members, and, independent from these functions, can act as a pro-survival factor (Morris et al 2011;Liu et al 2011;Strainic et al 2008;Ulfman et al 2005), we next examined whether C5a could have a similar function in hPSC. We therefore treated H9 hESC with 1nM C5a, or vehicle alone, 1 hour prior to enzymatic single cell dissociation and quantified cell survival of these single cell dissociated cultures after 16 hours.…”
