Differences in hippocampal protein levels between C57Bl/6J, PWD/PhJ, and Apodemus sylvaticus are paralleled by differences in spatial memory

Abstract: There is a significant strain-dependent performance in the Morris water maze (MWM), a paradigm for the evaluation of spatial memory. In contrast, information on molecular differences that may be responsible for differences in spatial memory performance is limited. The aim of the study was therefore to investigate differences in hippocampal protein levels in three groups with different performance in the MWM. C57BL/6J (inbred laboratory strain), PWD/PhJ (inbred strain derived from wild animals of Mus musculus),… Show more

“…This may be due to the reported strain-dependent protein expressional patterns [40][41][42]. The finding that among the many proteins only PP-1A was paralleling OM may be due to stringent statistics used for the gel-based proteomics approach.…”

“…Wild derived inbred strains possess many more genetic polymorphisms (Salcedo et al 2007), rapid decay of linkage disequilibrium (Laurie et al 2007) for high precision, and tremendous behavioral variation (Fernandes et al 2004; McShane et al 2010; Patil et al 2011). While some constitute separate sub-species, many can be intercrossed with common inbred strains and with each other (Forejt 1985).…”

“…Protein identification with nano-LC-ESI-CID/ETD-MS/MS was performed as described (Patil et al, 2010a). Protein spots of interest (17 spots from yoked vs. trained mice and 7 protein spots from the group good vs. 7 no-performers) were excised and put into a 0.5 ml protein lobind tube (Eppendorf, Germany).…”

“…A gel-based proteomics technique that was successfully used in several publications (Sunyer et al, 2008b;Zheng et al, 2009;Patil et al, 2010a) was applied and unambiguously identified differential protein levels in hippocampi.…”
