Diethyldithiocarbamate (DDC), a potent radical scavenger 1) and nitric oxide synthase inhibitor, 2) has been shown to inhibit lipid peroxidation and hydroxyl radicals in rats, 3,4) and has an anti-cataract effect in selenite-treated rats. 5,6) However, DDC is unstable in neutral solution, and is not able penetrate through the cornea into the aqueous humor.7) Disulfiram (DSF), a dimer of DDC, has long been used to treat alcoholic syndrome without severe side effects.8) DSF is a powerful antioxidant that scavenges reactive oxygen species such as hydroxyl radical, superoxide anion, and peroxynitrite, and chelates metal ions in rat eyes 9,10) ; DSF, as a prodrug of DDC, is an anticipated anti-cataract drug. 7) We previously demonstrated that the oral administration of DSF has an anti-cataract effect on selenite-treated rats.7) However, its application in the ophthalmic field is limited due to its poor water solubility.One problem with lipophilic drugs, such as DSF, administered as aqueous eye drops is obtaining the desired drug concentration in any ophthalmic drug delivery system. Cyclodextrins are cyclic oligosaccharides comprising R-D-glucose linked by R-(1-4) glucosidic bonds. The potential use of natural cyclodextrins and their synthetic derivatives has been extensively studied to improve certain properties of these drugs, such as their solubility, stability, and/or bioavailability.11) 2-Hydroxypropyl-b-cyclodextrin (HPbCD) is a cyclic oligosaccharide with a hydrophilic outer surface and a lipophilic cavity at its centre, and is capable of forming inclusion complexes with many lipophilic drugs by taking up the drug molecule, or part of it, into its lipophilic cavity. 12,13) In aqueous solution, hydroxypropylmethylcellulose (HPMC), a water-soluble polymer, increases the solubilizing effect of cyclodextrins on lipophilic drugs by increasing the stability constants of the drug/cyclodextrin inclusions.6) In a previous study, we prepared HPbCD solution containing DSF and HPMC (DSF eye drops), and found the instillation of the DSF eye drops delayed lens opacification in ICR/f rats.
14)Therefore, the application of DSF eye drops as an anticataract drug is expected.In this study for the development of anti-cataract drugs, the selection of the experimental animal is very important. The ICR/f rat is a recessive-type hereditary cataractous strain. Opacity in the lens usually becomes evident at around 75 d of age, and the appearance of cataracts in adult ICR/f rats is 100%.15) Previous investigations have revealed that the proteolyses of some crystallins are significantly enhanced in ICR/f rat lenses. 15,16) The Ca 2ϩ content in the cataractous lenses of ICR/f rats is about 10-fold higher than in the lenses of Wistar rats, and the autolytic products formed by calpain can also be detected in cataractous lenses. 17,18) It is noteworthy that ICR/f rat cataracts are not related to diabetic cataracts, and the mechanism of cataract development is similar to that of senile cataracts.
