Diencephalic-mesencephalic Junction Dysplasia: A Case Report and Overview of What is Known so far

Patricia Piazza Rafful,
Mareen Sarah Kraus

Abstract: Background: Diencephalic-mesencephalic junction dysplasia (DMJD) is a rare and recently newly described forebrain-midbrain malformation involving the upper aspect of the brainstem and resulting in an abnormal regionalization of the diencephalon and mesencephalon level. The fingerprint of DMJD is the pathognomonic butterfly-like appearance of the midbrain seen on the axial plane of brain magnetic resonance imaging (MRI). Initially, only two types of diencephalon-mesencephalon continuity were defined: type A des… Show more

“…The ganglionic eminence arising from the ventral telencephalon is an important transitory fetal brain structure that generates the basal ganglia, thalami, olfactory bulb, and cortical interneurons [4][5][6]. It is well known that Fgf8 that is expressed in the isthmus organizer at the boundary between the midbrain and hindbrain is a master regulator of the signaling cascade for early brain development, which drives expression of the engrailed (En) and Paired box (Pax) transcription factors and plays an essential role in signal transduction processes required for DMJ formation [1,7,8].…”

“…It is also postulated that L1CAM encoding a cell adhesion molecule and VRK1 encoding a serine/threonine kinase are responsible for rare cases with DMJD [3]. These cell adhesion molecules, transcription factor, and signal transduction kinase play a pivotal role in brain embryogenesis in conjunction with the FGF8signaling cascade [1,7,8].…”
