Diatrack particle tracking software: Review of applications and performance evaluation

Abstract: Object tracking is an instrumental tool supporting studies of cellular trafficking. There are three challenges in object tracking: the identification of targets; the precise determination of their position and boundaries, and the assembly of correct trajectories. This last challenge is particularly relevant when dealing with densely populated images with low signal-to-noise ratios - conditions that are often encountered in applications such as organelle tracking, virus particle tracking, or single-molecule ima… Show more

“…Aligned EB1 and CENP-A datasets were processed in three sequential steps using the Diatrack software package ( Vallotton et al, 2017 ). First, images were background subtracted and Gaussian filtered with a sigma of 1 pixel to reduce image noise.…”

“…The peripheral NS5A foci (which are putative replication complexes) are frequently highly motile and capable of rapid long-range traffic. Hence, advanced models of NS5A will need to incorporate and addresses the motile populations of NS5A foci, for which particle tracking analysis will be an important evaluation tool, cf, e.g., [ 39 ].…”

“…We took advantage of the observation that single NPCs can occasionally be detected in dividing yeast cells where the part of the NE bridging the mother and daughter nuclei elongates and narrows (Movie S1) (36). These NPCs are more separated from their neighbors, allowing us to image and quantify the intensity of individual NPCs in several thousand cells using the Diatrack particle tracking software (37). As an absolute intensity reference, we also imaged in parallel, yeastexpressed rotavirus-like VP2 particles that contain exactly 120 copies of yEGFP (38).…”
