Diapause-Associated Protein3 Functions as Cu/Zn Superoxide Dismutase in the Chinese Oak Silkworm (Antheraea pernyi)

Abstract: To better understand the molecular mechanism underlying of diapause in Antheraea pernyi (A.pernyi), we cloned a novel diapause-associated protein 3 (DAP3) gene from A.pernyi by reverse transcription-polymerase chain reaction (RT-PCR) and studied the biological functions. Sequence analysis revealed that this gene encodes 171 amino acids and has a conserved domain of Copper/Zinc superoxide dismutase (Cu/Zn-SOD). Western blot and qRT-PCR results showed that DAP3 was mainly expressed in the pupal stage, and gradua… Show more

“…We found strong down-regulation of SOD1-vtg fusion in response to osmotic (NaCl) and oxidative stress (UV-B light), whereas we observed up-regulation of SOD-vtg under Cd, carbaryl and both microcystin treatments (Table S1). SOD-vtg is generally expressed in diapausing insects (Bi et al 2014) and crustaceans (Acton 2012) ); hence, the regulation of this gene under environmental perturbation indicates a potential alteration of the diapause process by environmental perturbations (e.g. Slusarczyk & Rybicka 2011).…”

“…minax , a microsomal GST (mGST, BmUnigene15924.co) and a Mn-SOD (BmUnigene23806.co) were up-regulated throughout diapause ( S5 Fig ). Likewise, members of the GST or SOD family were also up-regulated in other insects during diapause [ 32 , 56 , 57 ]. The up-regulation of these antioxidant proteins is speculated to protect individuals from oxidative damage in hypoxia/anoxia environments.…”

“…In insects, SOD1-null flies show markedly decreased lifespans and are highly susceptible to oxidation [ 27 ]. Diapause-associated protein 3 has a copper/zinc superoxide dismutase (Cu/Zn-SOD) domain and is involved in the regulation of diapause in Antheraea pernyi [ 28 ]. Hyphantria cunea SOD1 and SOD2 play a role in resistance to oxidative stress, and SOD2, in particular, plays a role with apo lipophorin III in apoptosis [ 29 , 30 ].…”
