Diamine oxidase is involved in H2O2 production in the chalazal cells during barley grain filling

Abstract: The localization and activities of diamine oxidase (DAO, EC 1.4.3.6) and polyamine oxidase (PAO, EC 1.4.3.4) together with polyamine levels have been investigated in developing grains of barley (Hordeum vulgare L.). DAO (pH 7.5) is present mainly in vascular tissue and its neighbouring cells, namely chalazal cells and nucellar projection, while PAO (pH 6.0) is mainly localized in the chlorenchymatous cells of the crease and at the base of the vascular tissue. Activities of both these enzymes appear to be indep… Show more

“…Hydrogen peroxide is suggested to participate in lignin deposition in the cell walls in a peroxidase-catalyzed reaction [100]. The involvement of a diamine oxidase in H 2 O 2 production has been demonstrated along with lignin deposition in the chalazal cells, in developing barley grains, in developing barley grains [100].…”

Production of ROS and their release in the surrounding medium are supposed to play a part in protecting the embryo against pathogens during seed imbibition [99].…”

“…In suberin-and lignin-rich cell walls, the cross-linking of phenolics is mediated by H 2 O 2 (Fry, 1998;Schweikert et al, 2000), a process that occurs in chalazal cells during the final stage of barley grain development (Cochrane, 2000;Asthir et al, 2002). ROS are intimately involved in PCD (Apel and Hirt, 2004;Van Breusegem et al, 2008).…”

“…The supernatant was used as source of enzyme. DAO (EC 1.4.3.6) and PAO (EC 1.5.3.11) activities were assayed as described by Asthir et al (2002) using Put (for DAO) and Spd (for PAO) as substrates. Activities are expressed as nmol (D-pyrroline) min -1 g -1 FW for DAO and PAO.…”
