Background Tubal pregnancy is recognized as one of the most common ectopic pregnancy types. Salpingitis may result in tubal pregnancy by causing fallopian tube occlusion and hydrosalpinx. B cell activation factor (BAFF) is a proinflammatory cytokine that helps regulate both innate and adaptive immune responses. Our previous study firstly showed that BAFF immunostaining appeared on the cellular membrane and in the cytoplasm of tubal epithelial cells, and both BAFF protein and mRNA in human inflamed fallopian tubes had higher expression levels than those in normal fallopian tubes. This study aimed to elucidate the association between the expression of BAFF gene and the inflammation in the human fallopian tube leading to tubal pregnancy. Methods We examined 70 patients undergoing salpingectomy for salpingitis ( n = 35) and tubal pregnancy ( n = 35). Twenty patients with benign uterine diseases undergoing complete hysterectomy and salpingectomy were recruited into control group. BAFF mRNA and protein in tissue samples were detected by qPCR and Western blotting methods. Furthermore, serum levels of BAFF, tumor necrosis factor-α (TNF-α) and interleukin (IL)-6 were measured using ELISA kits. Results We found statistically significantly elevated expressions of BAFF mRNA or protein in whole tissue samples, and serum levels of BAFF, TNF-α and IL-6 in whole blood samples from patients with salpingitis and tubal pregnancy, in comparison to the control group. Conclusion Based on the results, high expression of BAFF gene might induce inflammation in the human fallopian tube, suggesting its possible role in the tubal pregnancy process.

Cytokines, including interleukins, interferons, tumor necrosis factors, and chemokines, have a variety of pro- and anti-inflammatory effects in the body through a number of biochemical pathways and interactions. Stimuli, actions, interactions, and downstream effects of cytokines have been investigated in more depth in recent years, and clinical research has also been conducted to implicate cytokines in causal patterns in certain diseases. However, particular cutoffs of cytokines as biomarkers for disease processes have not been well studied, and this warrants future work to potentially improve diagnoses for diseases with inflammatory markers. A limited number of studies in this area are reviewed, considering diseases correlated with abnormal cytokine profiles, as well as specific cutoffs at which cytokines have been deemed clinically useful for diagnosing those diseases through Receiver Operator Characteristics modeling. In light of studies such as those discussed in this review, cytokine testing has the potential to support diagnosis due to its lack of invasiveness and low cost, compared to other common types of testing for infections and inflammatory diseases.

Mean±SD Serum concentration levels of IL1RA for ectopic pregnancy and controls group were (9.3375±3.260ng/ml), (2.1151±1.16ng/ml) respectively and this difference was highly significant (P value 0.00), while IL2 concentration Mean±SD in ectopic pregnancy was (2.2582±1.224ng/ml) and control group was (4.9923±1.59ng/ml) (P<0.05) with no statistically significance, Serum concentration level of IL6 for ectopic pregnancy was (23.2508±7.18 Pg/ml) while controls group was (9.0025±2.74 Pg/ml) with statistically significant differences (P value 0.00), IL8 level in ectopic pregnancy and controls show (76.9892±19.8 Pg/ml), (26.5498±11.85 Pg/ml) respectively and this different were statistically significant (P value 0.00).Serum concentration levels of TNFα for ectopic pregnancy and controls group were (31.5328±10.22 Pg/ml), (9.5346±10.22 Pg/ml) respectively this different was statistic significant (P value 0.00).
