Diagnoses of fipronil resistance in Brazilian cattle ticks (Rhipicephalus (Boophilus) microplus) using in vitro larval bioassays

Abstract: Fipronil is a phenylpyrazolic insecticide that is widely used in agriculture and has been recently used to control the cattle tick, Rhipicephalus (Boophilus) microplus. Because of the serious problems associated with resistance to the available acaricides, this product has been used as an important alternative to control acaricide-resistant ticks. The objective of this work was to analyse the fipronil sensitivity of ticks that were collected from farms with a history of fipronil use by larval bioassays. A tota… Show more

“…microplus to the fluazuron pharmacological basis, but attention must be paid to the fact that if any product is not applied to the animals in an appropriate way and at a suitable time, the resistance will tend to appear. Regarding ivermectin, reports described resistance in Mexico (PEREZ-COGOLLO et al, 2010) and the field experiment reported by Martins and Furlong (2001) discovered a strain resistant to avermectins in the State of Rio Grande do Sul; also, this same situation was already described in reports in the states of São Paulo and Mato Grosso do Sul (CASTRO-JANER et al, 2010b).…”

“…In Uruguay (CASTRO-JANER et al, 2010a), in the Brazilian states of Rio Grande do Sul, Mato Grosso do Sul and São Paulo, the existence of fipronil resistant strains (CASTRO-JANER et al, 2010b) was also shown. This situation points to the fact that this drug must be carefully used by Bahia State herd owners, since the inadequate application can lead to the development of resistant ticks.…”

“…The LIT was described by Shaw (1966) and several adaptations and modifications have been made over the years. Although this assay has not yet been recognized by FAO as a trustworthy method for detecting acaricide resistance in tick populations, it has been used to assess the toxicity of chemicals that act systemically, such as fipronil (CASTRO-JANER et al, 2010) and ivermectin (KLAFKE et al, 2006(KLAFKE et al, , 2012. Recently, it was described as a reliable method for detecting resistance to amitraz (MENDES et al, 2013).…”

“…(B.) microplus can be diagnosed using the larval stage in in vitro bioassays: a) larval packet test (LPT), originally described by Stone and Haydock (1962), standardized by FAO (1971) and then modified by Miller et al (2002), with the aim of specifically diagnosing amitraz resistance; b) larval immersion test (LIT), originally described by Shaw (1966), then adapted for microcentrifuge tubes by Sabatini et al (2001) and used successfully for diagnosing resistance to ivermectin (KLAFKE et al 2006(KLAFKE et al , 2012 and fipronil (CASTRO-JANER et al, 2010); and c) syringe immersion test (SIT), described by Souza et al (2008) although not yet standardized for routine use as an acaricide resistance diagnostic tool. Recently, Mendes et al (2013) described the use of LIT for diagnosing amitraz resistance in R.…”
