Diabetic modifier QTLs in F2 intercrosses carrying homozygous transgene of TGF-β

Abstract: When the homozygous active form of porcine TGF-beta1 transgene (Tgf/Tgf) (under control of the rat glucagon promoter) is introduced into the nonobese diabetic mouse (NOD) genetic background, the mice develop endocrine and exocrine pancreatic hypoplasia, low serum insulin concentrations, and impaired glucose tolerance. To identify genetic modifiers of the diabetic phenotypes, we crossed hemizygous NOD-Tgf with DBA/2J mice (D2) or C3H/HeJ mice (C3H) and used the "transgenic mice" for quantitative trait loci (QTL… Show more

“…This gene encodes a member of the vitamin B12-binding protein family, cobalamin metabolic process, which is found in neutrophilic granules. TCN1 has shown linkage to serum insulin concentrations in impaired glucose tolerance [69]. TCN1 protein was also increased in the synovium of rheumatoid arthritis [70] and was significantly associated with cholesterol levels or statin response [71], which may provide a predisposing link between skin inflammation and high cholesterol.…”

“…Therefore due to the implication of the identified genes at the early pre-inflammatory stages of T1D it was interesting to search for the presence of variants, by referring to the existence of the available unique single nucleotide polymorphic loci (SNPs) of the mouse genome ( Frazer et al, 2007 ). We compared strains that were used as mating partners in crosses with the NOD mouse in genetic studies ( Leiter et al, 1987 , McClive et al, 1994 , Peterson et al, 1994 , Suzuki et al, 2008 ) and assessed for polymorphisms between these strains (C57Bl/6, C3H, DBA/2, SJL/J and NON/ Lt J), with as reference, the NOD/ ShiLt J.…”
