There is recently a fast growing interest in green nanotechnology as a better approach for managing diabetes mellitus (DM) and its associated complications. This study therefore aimed at exploring the protective potentials of biogenic phytomediated magnesium-based nanoparticles using Monodora myristica seed (Mg(OH)2NP-Mm) against streptozotocin-induced diabetic hepatopathy, nephropathy and neuropathy in Wistar rats. Mg(OH)2NP-Mm was biosynthesized and characterized using standard techniques. Forty-eight (48) adult male Wistar rats weighing 150-200 g, were indiscriminately grouped into eight (8) groups of six (6) rats (n=6) each. Diabetes was induced with a low dose of streptozotocin (STZ) (55 mg/kg bw) and diabetic animals administered 50, 100, 150 and 200 mg/kg bw Mg(OH)2NP-Mm for 21 days, while control groups received glibenclamide (5 mg/kg bw) and Mg(OH)2-STD (150 mg/kg bw), respectively. In this study, treatment with Mg(OH)2NP-Mm caused a significant (p < 0.05) improvement in fasting blood sugar (FBG), serum hepatic biomarkers (AST, ALT and ALP activities), renal clearance markers (creatinine and urea), total protein (TP) and bilirubin. Enzymatic and non-enzymatic antioxidants, as well as histomorphological examinations indicated a significant (p < 0.05) restoration of the hepatic, renal and brain tissues. This finding therefore, hypothesizes attenuation of redox imbalance which probably could be the basis for the protective effect demonstrated by Mg(OH)2NP-Mm in the tissues examined from STZ-induced diabetic rats.

Identification of Up-Regulated ANXA3 Resulting in Fracture Non-Union in Patients With T2DM

Diabetes mellitus is a metabolic disorder that increases fracture risk and interferes with bone formation and impairs fracture healing. Genomic studies on diabetes and fracture healing are lacking. We used a weighted co-expression network analysis (WGCNA) method to identify susceptibility modules and hub genes associated with T2DM and fracture healing. First, we downloaded the GSE95849, GSE93213, GSE93215, and GSE142786 data from the Gene Expression Omnibus (GEO) website, analyzed differential expression genes and constructed a WGCNA network. Second, we screened out 30 hub genes, which were found to be enriched in neutrophil activation, translational initiation, RAGE receptor binding, propanoate metabolism, and other pathways through Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG), and gene set enrichment analysis (GSEA) analyses. Third, we searched for genes related to bone metabolism and fracture healing in the published genome-wide single nucleotide polymorphism (SNP) data, built a protein-protein interaction (PPI) network with hub genes, and found that they were associated with metabolic process, blood vessel development, and extracellular matrix organization. ANXA3 was identified as the biomarker based on gene expression and correlation analysis. And the AUC value of it was 0.947. Fourth, we explored that ANXA3 was associated with neutrophils in fracture healing process by single-cell RNA sequencing analysis. Finally, we collected clinical patient samples and verified the expression of ANXA3 by qRT-PCR in patents with T2DM and fracture non-union. In conclusion, this is the first genomics study on the effect of T2DM on fracture healing. Our study identified some characteristic modules and hub genes in the etiology of T2DM-associated fracture non-union, which may help to further investigate the molecular mechanisms. Up-regulated ANXA3 potentially contributed to fracture non-union in T2DM by mediating neutrophils. It can be a prognostic biomarker and potential therapeutic target.

Herbal medication developed from natural resources has to have antibacterial and antioxidant effects. The aim of this research is to look at the chemical makeup of Solanecio gigas (S. gigas) stem bark essential oil (EO), as well as the effectiveness of EO and extracts (chloroform, ethyl acetate, and methanol) against human pathogenic bacteria and their antioxidant activity. The GC-MS analysis identified 23 components, accounting for 98.7% of the total oil containing Methylene chloride (49.2%), sabinene (10.5%), 1-nonene (11.3%), Terpinen-4-ol (6.9%), Camphene (4.3%), γ-terpinene (3.6%), α-phellandrene (2.9%) β-myrcene (2.6%), 1,2,5-Oxadiazol-3-carboxamide, 4,4′-azobis-2,2′-dioxide (2.4%), α-terpinene (1.9%), 1-Octanamine, N-methyl- (1.9%), ρ-cymene (1.6%) as major components. The antibacterial efficacy of the EO and extracts (25, 50, 100, and 200 mg/ml) was demonstrated by the inhibitory zones (8.5 ± 0.47–23.3 ± 0.36 and 7.2 ± 0.25–22.0 ± 0.45 mm), respectively. The MIC values of the extracts and the EO were 120–150 and 240 to <1100 μg/ml, respectively. The EO also demonstrated a significant antibacterial impact. The EO and methanolic extract had free radical scavenging activities with IC50 value, 13.8 ± 0.48 and 4.2 ± 0.04 μg/ml, respectively. In comparison to the other extracts, the methanolic extract had the greatest phenolics (100.2 ± 0.13 μg GAE/mg of dry extract) and flavonoid contents (112.1 ± 0.18 μg CE/mg of dry extract).
