Diabetes mellitus and cancer: a system of insulin-like growth factors

Abstract: Diabetes mellitus and malignant tumors are among the most common and complex diseases. Epidemiological studies have shown a strong relationship between these pathologies. The causality of this relationship has not yet been unambiguously established, but a number of probable biological mechanisms have been proposed to explain it through the effects of hyperglycemia, hyperinsulinemia on the process of oncogenesis. М. с соавт., подробно обсуждаются современные взгляды на значение системы инсулиноподобных факторов роста и, в частности, указывается, что злокачественному фенотипу в значительной степени способствуют нарушения регуляции оси инсулиноподобных факторов роста (IGF -Insulin-like growth factor 1) связывающих их белков (IGFBP -nsulin-like growth factor-binding protein) -изменение биодоступности лигандов инсулиноподобных факторов роста 1,2 (IGF-1,-2), сверхэкспрессия рецепторов к инсулиноподобному фактору роста 1 (IGF1R), нарушение регуляции нижестоящих сигнальных эффекторов. Хроническая гиперинсулинемия способствует увеличению уровня биодоступного IGF-1 (за счет увеличения уровня IGF-1 и снижения уровня связывающих белков IGFBP-1, -2, -3), а также увеличению образования гибридных рецепторов инсулина и IGF1R, тем самым увеличивая митогенный потенциал клетки [42]. Таким образом, снижая гиперинсулинемию, метформин демонстрирует непрямой противоопухолевый эффект.…”
