Dexmedetomidine Protects against Airway Inflammation and Airway Remodeling in a Murine Model of Chronic Asthma through TLR4/NF-κB Signaling Pathway

Abstract: Asthma is a common respiratory disease characterized by chronic airway inflammation. Dexmedetomidine (DEX), a highly selective α2 adrenergic receptor agonist, has been shown to participate in regulating inflammatory states and thus exert organ protective actions. However, the potential of DEX in asthma is still unknown. This study is aimed at investigating the role of DEX in a mouse model of house dust mite- (HDM-) induced asthma and exploring its underlying mechanism. Here, we found that DEX treatment signifi… Show more

“…Huperzin is a natural compound extracted from the root of Huperzia serrata with anti-inflammatory, antioxidant and antitumor effects. Zhou et al [33] proved by animal experimental studies that thujone glucoside can reduce the expression level of inflammatory factors TNF-α and IL-6, and inhibit the expression of PI3K, AKT, and mTOR proteins in thujone glucoside group of rats, which can prove that thujone glucoside can inhibit the PI3K/ AKT/mTOR pathway in COPD rats, inhibit the inflammation in the airway, and play a protective role against lung injury.…”

“…Hypoxemia occurs more frequently during one-lung ventilation, and the degree of ischemia and hypoxia increases as the duration of single lung ventilation exceeds 3 hours. Lung injuries caused by the single lung ventilation can bring secondary harm to patients who have mild-to-moderate abnormal lung function before surgery, and severe cases can eventually develop into acute lung injury ( 11 , 12 ). Moreover, surgical treatment may lead to adverse stress reactions such as activation of inflammatory reactions, limitation of immune functions, and an increase in the risks of tumor recurrence and metastasis.…”

“…These molecules activate RA-fibroblast-like synoviocytes (RA-FLSs) through regulation of proliferation, migration to synovial tissue, and inflammatory reactions [1]. Dexmedetomidine (Dex), which is known as a pain reliever and activator of the α2-adrenergic receptor (α2-AR), attenuates the cAMP level [4,5] and plays an anti-inflammatory role by inhibiting the NF-κB [6][7][8] and Janus kinase 2 (JAK2)-signal transducer and activator of transcription 3 (STAT3) signaling pathways [9][10][11]. Dex has been reported to play an inhibitory role in various inflammation-associated diseases, such as RA, and has thus been reviewed [12].…”
