Dexamethasone Induces an Acute and Sustained Rise in Circulating Leptin Levels in Normal Human Subjects

Abstract: Leptin, the protein product of the ob gene, is thought to have a role in signalling satiety through hypothalamic pathways. Glucocorticoids are potent stimulators of both ob gene expression and circulating leptin levels in the rat, yet are powerful appetite stimulants in humans. We have investigated circulating leptin responses to intermediate term and acute administration of dexamethasone. Dexamethasone 2 mg twice daily resulted in a rapid and sustained rise in 08.00 h leptin levels from basal values of 1.36 +… Show more

“…This response of leptin is opposing to the sensitized surge of corticosterone in the same animals, which has been considered as a pathophysiological feature of hyperactive HPA axis in human depression [8]. However, the stress-induced fall in leptin concentrations is unlikely to be mediated by corticosterone, as glucocorticoids have been shown to stimulate, rather than to inhibit, leptin production and release [9][10][11].…”

“…Individuals with any other condition that may possibly influence leptin or ghrelin levels, such as chronic inflammatory intestinal diseases or rheumatoid disorders, other chronic inflammatory diseases, malignant tumours, dysfunction of the thyroid gland, diabetes mellitus and cardiac disease (New York Heart Association Functional Class oII ), as well as treatment with corticosteroids [23][24][25], sex hormones [26] and b-blockers [27] were excluded from both groups, as were longterm fasting individuals [28]. Since ghrelin is a stomachderived hormone, patients with gastric disease or prior gastric surgery were excluded from the study.…”

“…In most in vitro and in vivo studies, glucocorticoids seem to upregulate leptin secretion and serum leptin levels (22)(23)(24). A minor inhibitory effect of dexamethasone (DEX) in a mouse cell line (14), as well as unchanged serum leptin levels after high doses of prednisolone given to healthy volunteers, have also been reported (25).…”
