Dexamethasone effects on creatine kinase activity and insulin‐like growth factor receptors in cultured muscle cells

Abstract: We examined the effects of dexamethasone on creatine kinase (CK) activity and insulin-like growth factor I (IGF-I) binding in two skeletal muscle-derived cell lines (mouse, C2C12; rat, L6) and in one cardiac muscle-derived cell line (rat, H9c2). Dexamethasone treatment during differentiation of cultured cells caused a dose-dependent increase in CK activity as well as an increase in the degree of myotube formation in C2C12 and L6, whereas H9c2 cells did not exhibit significant CK activities during culture or de… Show more

“…The LDH and citrate synthase activity increase at differentiation in the latter cell line. CK activity measured on serumderived C2C1~ myotubes was higher than found in a previous study (Whitson et al, 1989). They reported a twofold higher CK activity when C2C12 cells were cultured for 8 d on 2% horse serum with at least 3 ~rM dexamethasone.…”

“…A large proportion of the myoblasts did not fuse. These cells may reflect the quiescent satellite-like reserve cells as described for CzC12 mouse myoblast cells (Yoshida et al, 1998). When subcloned, these reserve cells retain their capacity to generate cells that fuse into myotubes.…”

“…For AIB uptake, this was associated with an increase in type I IGF receptor number. In addition, dexamethasone has recently been shown to potentiate the effect of IGF-I on creatine ki- nase activity in myogenic cells (Whitson et al, 1989). In contrast, we found no effect of dexamethasone on either basal or IGF-I stimulated AIB uptake in chicken myotubes (Fig.…”

“…Dexamethasone induces glutamine synthetase in L6 myoblasts, with a significant decrease of total protein synthesis (Max et al, 1987). Moreover, its interference with growth factor circuits has been documented in different normal myogenic models (Whitson et al, 1989;Southorn & Palmer, 1990;Poon et al, 1991).…”
