“…The consistency of typing results was evaluated considering previously typing through already validated kits such as AmpF l STR ® Identifiler Plus (7), AmpF l STR ® NGM PCR kits (Life Technologies, Carlsbad, CA, USA), PowerPlex ® 16 HS System (8) or PowerPlex ® ESX 17 System (9) (Promega, Madison, WI, USA). DNA concentration was determined by measuring the absorbance of the sample at 260 nm, using the spectrophotometer NanoDrop 1000 (ThermoScientific, Waltham, Massachusetts, USA).…”

“…Two simulated reference control samples M and O were obtained from swabbing the inside of the cheek of two individuals and transferred to FTA® paper. Samples M and O were also extracted using phenol‐chloroform‐isoamyl alcohol and amplified using PowerPlex16 HS (Promega) under the manufacturer's standard conditions and used as comparison samples to test extraction efficiency . A small reference population was also obtained from 18 individuals by swabbing the inside of their cheeks and transferring the swabbing to the FTA® paper; these samples were then analyzed.…”

“…These analyses are conducted through the use of commercially-available kits, such as Identifiler 1 Plus (Applied Biosystems 1 , Foster City, CA) and the PowerPlex 1 16HS System (Promega Corporation, Madison, WI). These kits are designed to co-amplify targets of 15 polymorphic STR loci plus Amelogenin, a sex determining marker, and contain advanced buffering components designed to produce results in the presence of polymerase chain reaction (PCR) inhibitors [1,2]. Optimal performance of these kits requires adding DNA template to PCR reagents within the working range of the kit.…”

“…Taking into consideration the potential challenges of attempting to quantify inhibited DNA samples, two questions are addressed: (1) Are DNA samples that include metal ions accurately quantified? and, (2) Does the IPC detect the presence of metal ion PCR inhibitors? Because metal ions have the potential to interact directly with DNA and inhibit PCR, it is important to elucidate the reliability of results obtained from PCR-based methods in their presence and the ability of the IPC to differentiate a true negative from an inhibited sample.…”
