Developmental exposure to lead (Pb) alters the expression of the human tau gene and its products in a transgenic animal model

Abstract: Tauopathies are a class of neurodegenerative diseases associated with the pathological aggregationof the tau protein in the human brain. The best known of these illnesses is Alzheimer's disease (AD); a disease where the microtubule associated protein tau (MAPT) becomes hyperphosphorylated (lowering its binding affinity to microtubules) and aggregates within neurons in the form of neurofibrillary tangles (NFTs). In this paper we examine whether environmental factors play a significant role in tau pathogenesis. … Show more

“…Exposure of wild-type mice to lead-contaminated drinking water at birth resulted in increased levels of hyperphosphorylated tau in vivo when the mice were old [ 77 ]. Lead-mediated tau hyperphosphorylation is driven by the increased activity of the CDK5/p25 complex as well as the GSK-3β kinase [ 77 , 78 , 79 , 80 ]. Altogether, these findings highlight that lead not only induces Aβ aggregation by increasing APP cleavage activity but also induces tau aggregation via the CDK5/p25 complex and GSK-3β kinase.…”

“…This demonstrates that early exposure to pollutants induces effects observable in older animals. Early exposure to lead induced a similar cognitive decline, neuroinflammation [111, 112], A β peptide overproduction [113, 114], and tau hyperphosphorylation [115118] in both mice and rats. Interestingly, Bihaqi et al [119] observed a cognitive decline in rats only when exposed to lead as pups not as adults.…”

“…The in vivo studies in rat model revealed that Pb exposure leads to the increased amount and to hyperphosphorylation of Tau protein [167]. Enhanced tauopathy in animal models has also been reported by other groups [168,169]. Pb has been also proven to increase amyloid β and senile plaques formation in transgenic mice and in monkey model [170,171].…”
