Abstract. The purpose of this study was to investigate fertilization ability and embryo development to the blastocyst stage after reciprocal in vitro fertilization (IVF) between yak and cattle in an attempt to clarify the problem of low conception rate after mating yak females with cattle bulls. In vitro-matured (IVM) cattle and yak oocytes were inseminated with either Holstein or yak spermatozoa, and after an 18-h of coincubation period, a proportion of the oocytes was fixed and examined for sperm penetration, polyspermy and male pronuclear formation. The remaining oocytes were cultured in vitro and evaluated for cleavage and blastocyst formation rates. The percentage of IVM oocytes penetrated by spermatozoa ranged from 78.5 to 90.5%, and the formation of one or two pronuclei and the incidence of polyspermy did not differ among the different combinations. The cleavage and blastocyst rates were not affected by the species of the sperm, but they were affected by the species of the oocytes (P<0.05), with cattle oocytes having a higher (P<0.05) cleavage and blastocyst rates (69.9 and 31.3%) than yak oocytes (62.7 and 11.5%). The blastocyst formation rate was calculated from the cleaved zygotes. The interaction between sire and oocytes species (P<0.05) influenced blastocyst formation rate, with the highest blastocyst rate occurring in cattle oocytes fertilized with yak spermatozoa (36.5%) and the lowest rate occurring in yak oocytes fertilized with yak spermatozoa (9.4%). The effect of heterosis was apparent at the blastocyst stage, but there was a large reciprocal difference in blastocyst production between crosses. It was concluded that the low conception rate that results from crossing yaks with cattle is not due to either a speciesspecific block of fertilization or the developmental competence of the early stage embryo. Dev. 55: [480][481][482][483] 2009) he yak is one of the world's most remarkable domestic animals -a herbivore living on the "roof of the world", in and around the Himalayas and further north at altitudes ranging from 2500 to 5500 m with no frost-free period and mostly above the tree line. They are very important to local people for meat, milk and draft power given that few other animals survive in these areas. However, their production traits are inferior to those of improved cattle (Bos taurus) breeds [1,2].Interest in the commercial use of local cattle × yak hybrids has existed for at least 3000 years. During this time, attempts have been made to improve meat and milk production (quoted from Wiener et al. [1]). A number of systematic studies carried out from 1950s to 1990s have examined the meat and milk performance of hybrids of yak derived from both natural mating and artificial insemination using semen from exotic breeds of cattle such as the Holstein, Simmental, Hereford and Shorthorn. Resumo -O objetivo deste trabalho foi avaliar os efeitos da doadora, estação do ano, do touro e do tipo de sêmen, na produção in vitro de embriões da raça Sindi (Bos indicus). Para isso, avaliaram-se 434 sessões de aspiração folicular in vivo (OPU), realizadas em 152 doadoras, com a utilização de sêmen sexado e não sexado de 22 touros. Analisaram-se as seguintes variáveis: doadora; idade da doadora; estação do ano; touro; tipo de sêmen; oócitos recuperados; viáveis e degenerados; e as taxas de clivagem e de blastocistos. Observaram-se efeitos da doadora e do touro sobre os oócitos recuperados, viáveis e degenerados e sobre a taxa de blastocisto. O ano foi dividido em estações chuvosa, de outubro a março, e seca, de abril a setembro. Na estação chuvosa, a proporção de oócitos viáveis aumentou, enquanto a de degenerados diminuiu. As doadoras com menos de seis anos tiveram maior proporção de oócitos viáveis e menor, de degenerados. O sêmen não sexado obteve as melhores taxas de clivagem e de blastocistos. Doadoras de até seis anos apresentam a maior proporção de oócitos viáveis, e o maior número de oócitos viáveis é produzido na estação chuvosa. To this end, 434 in vivo ovum pick up (OPU) sessions, performed in 152 oocyte donors, were evaluated using sex-sorted or unsorted semen from 22 sires. The following variables were analyzed: donor; age of the donor; season of the year; sire; type of semen; recovered, viable, and degenerated oocytes; and cleavage and blastocyst rates. The effects of the donor and sire were observed on recovered, viable, and degenerated oocytes, and on blastocyst rates. The year was split into rainy season, from October to March, and dry season, from April to September. In the rainy season, the proportion of viable oocytes increased, while the proportion of the degenerated ones reduced. Donors with less than six years of age had a higher proportion of viable oocytes and a lower proportion of degenerated ones. Unsorted semen obtained the best cleavage and blastocyst rates. Donors with up to six years of age show the greatest proportion of viable oocytes, and the greatest number of viable oocytes is produced during the rainy season. Unsorted semen shows the best rates for cleavage and blastocysts.

-The objective of this work was to evaluate the effects of hormonal synchronization protocols, associated or not with follicular development stimulation, on the recovery of oocytes and on in vitro production of Bos indicus and B. taurus embryos, in different seasons. Ultrasound-guided follicular aspirations (n=237) were performed without pre-treatment (G1, control group) and after follicular wave synchronization (G2), or after follicular wave synchronization and follicle growth induction (G3). Bos indicus produced more oocytes and embryos than B. taurus (18.7±0.9 vs. 11.9±0.6 oocytes and 4.8±0.3 vs. 2.1±0.2 embryos). On average, oocyte and embryo yields were higher in G3 than in G2, and both were greater than in G1, which lead to a higher conversion of oocytes to embryos in these treatments. The hot or the cold season did not affect the B. indicus outcomes, whereas, in B. taurus, both oocyte recovery and embryo production were higher in the cold season. Follicular wave synchronization improves ovum pick-up and in vitro production of embryos in both cattle subspecies evaluated.Index terms: Bos indicus, Bos taurus, artificial insemination, follicle growth induction, follicular wave synchronization, FSH stimulation. As aspirações foliculares guiadas por ultrassom (n=237) foram realizadas sem pré-tratamento (G1, grupo controle) e após sincronização da onda folicular (G2), ou após sincronização da onda folicular e estímulo do desenvolvimento folicular (G3). Bos indicus produziu mais oócitos e embriões que B. taurus (18,7±0,9 vs. 11,9±0,6 oócitos e 4,8±0,3 vs. 2,1±0,2 embriões). Em média, as produções de oócitos e embriões foram maiores em G3 do que em G2, e ambas foram maiores que em G1, o que levou a uma maior conversão de oócitos em embriões nesses tratamentos. A estação quente ou fria não afetou os resultados de B. indicus, enquanto, em B. taurus, tanto a recuperação de oócitos quanto a produção de embriões foram maiores na estação fria.
