Effect of culture methods on cumulus and oocyte morphology and meiotic competence of bovine oocytes from early antral follicles* AbstractThe objective of the present study was to establish a culture system that would maintain the three-dimensional structure of bovine early antral follicles (EAF) or isolated cumulus-oocyte-granulosa complexes (COCGs) and increase the resulting portion of COCs with normal morphology for subsequent IVM. The morphological quality and meiotic competence of oocytes originating from early antral bovine ovarian follicles (0.2 to 0.7 mm and 0.4 to 0.7 mm diameter) were evaluated following culture in vitro for 14 and 7 d, respectively, and subsequent in vitro maturation. Growth culture modifications included culture in the well of the well (WOW) system; a microdroplet of collagen gel (2 x 5 µl vs. 2 x 400 µl; standard system) and culture of EAF in hanging drops of medium (inverted system). Significantly higher (P<0.01) proportions of COCs with normal morphology (60.4%) were obtained from COCGs compared to EAF (4.8%) grown in the WOW system. Embedding of COCGs in microdrops of collagen gel significantly increased proportion of COCs with normal morphology (63.2%) compared to those embedded in standard volume gels (35.3%). Recovery rate of COCs with normal morphology from cultured EAF was improved both by using microdrops of gel (44%) and by culture in the inverted system (39.3%) over that found for the standard system (8.5%).

This research has been designed to study the major events of nuclear remodeling that characterize sheep oocytes during the early stage of folliculogenesis (transition from preantral to antral stage). In particular, the modifications in large-scale chromatin configuration, the global DNA methylation, and the process of telomere elongation have been investigated as crucial events of oocyte nuclear maturity. In addition, the spatio-temporal distribution of the major enzymes involved in DNA methylation, the DNA methyltransferase 1 (Dnmt1), and in telomere elongation, telomerase catalytic subunit (TERT), have been described. To these aims, the nuclei of isolated oocytes were investigated using immunocytochemistry and Q-FISH analyses. In absence of preliminary information, these nuclear determinants were compared with those of fully competent germ cells obtained from medium and preovulatory antral follicles. The nuclei of sheep oocytes acquired a condensed chromatin configuration, stable high levels of global DNA methylation, and a definitive telomere length already in the majority of late growing stage oocytes (110 microm) derived from early antral follicles. In addition, while the process of methylation resulted strictly related to oocyte diameter, the telomeric program appeared to be highly chromatin configuration-dependent. The translocation of Dnmt1 and TERT from the nucleus to the cytoplasm in the oocytes derived from early antral follicles seems to confirm the definitive chromatin asset of these germ cells. In conclusion, changes in large-scale chromatin structure, epigenesis, and telomere size in the sheep are established prior to oocyte acquires the ability to resume meiosis.

This study assessed the histones methylation profile (H3K4me3 and H3K9me3) in late preantral (PA) and early antral (EA) caprine follicles grown in vivo and in vitro, and the anethole effect during in vitro culture of PA follicles. Uncultured in vivo–grown follicles (PA, n = 64; EA, n = 73) were used as controls to assess the methylation profile and genes' expression related to apoptosis cascade (BAX, proapoptotic; BCL2, antiapoptotic), steroidogenesis (CYP17, CYP19A1), and demethylation (KDM1AX1, KDM1AX2, KDM3A). The isolated PA follicles (n = 174) were cultured in vitro for 6 days in α‐MEM+ in either absence (control) or presence of anethole. After culture, EA follicles were evaluated for methylation, mRNA abundance, and morphometry. Follicle diameter increased after culture, regardless of treatment. The methylation profile and the mRNA abundance were similar between in vivo–grown PA and EA follicles. Anethole treatment led to higher H3K4me3 fluorescence intensity in EA follicles. The mRNA abundances of BAX, CYP17, and CYP19A1 were higher, and BCL2 and KDM3A were lower in in vitro–grown EA follicles than in vivo–grown follicles. In conclusion, in vitro follicle culture affected H3K4me3 fluorescence intensity, mRNA abundance of apoptotic genes, and steroidogenic and demethylase enzymes compared with in vivo–grown follicles.
