Development of Two Types of Rapid Diagnostic Test Kits To Detect the Hemagglutinin or Nucleoprotein of the Swine-Origin Pandemic Influenza A Virus H1N1

Abstract: Since its emergence in April 2009, pandemic influenza A virus H1N1 (H1N1 pdm), a new type of influenza A virus with a triple-reassortant genome, has spread throughout the world. Initial attempts to diagnose the infection in patients using immunochromatography (IC) relied on test kits developed for seasonal influenza A and B viruses, many of which proved significantly less sensitive to H1N1 pdm. Here, we prepared monoclonal antibodies that react with H1N1 pdm but not seasonal influenza A (H1N1 and H3N2) or B vi… Show more

“…With this pandemic virus of mixed-origin, increased anxiety levels resulted worldwide, accompanied by CDC guidelines suggesting prompt antiviral treatment and implementation of specific infection control precautions for hospitalized and high-risk patients [CDC, 2009a,c;Mizuike et al, 2011]. Under these circumstances, rapid diagnosis was pivotal for the initiation of treatment with antiviral drugs and management of infection control measures.…”

“…Mizuike et al developed an RIDT containing two mAbs specific to the HA and NP of H1N1pdm09 for the detection of H1N1pdm09 (Mizuike et al, 2011). Samuel et al (2011) generated mAbs to the HA1 of the H1N1pdm09 viral HA for the serodiagnosis of H1N1-pdm09 infections.…”

“…Consequently, we developed murine monoclonal antibodies that were specific for H1N1pdm, which exhibited no cross-reactivity with seasonal influenza A (H1N1 or H3N2) or seasonal influenza B viruses. These monoclonal antibodies were then employed to develop a new IC test kit for the high-sensitivity detection of H1N1pdm [27].…”
