Development of serotonin-induced ion currents in identified embryonic Retzius cells from the medicinal leech (Hirudo medicinalis)

Abstract: Retzius cells and the cutaneous baroreceptive P-cells of adult medicinal leeches (Hirudo medicinalis) respond to 5-HT by Cl- as well as by monovalent cation conductances (Drapeau et al., 1989). However, chemical synaptic connections between Retzius cells (Liu and Nicholls, 1989) as well as Retzius cells and P-cells in culture (Drapeau and Sanchez-Armass, 1988) are mediated by postsynaptic Cl- currents in response to 5-HT release from the presynaptic cell. It was the aim of the present study to find out whether… Show more

“…These E m changes are mainly due to the direct activation of 5-HT-gated Cl Ϫ channels (Lessmann and Dietzel, 1995b;Ali et al, 1998) as well as to the activation of unselective cation channels via metabotropic 5-HT receptors (Catarsi and Drapeau, 1997;Drapeau et al, 1989). Beside these well-characterized systems, the existence of other 5-HT-dependent conductances has been proposed (Goldermann et al, 1994;Lessmann and Dietzel, 1991;Walz and Schlue, 1982).…”

“…Our data suggest the hypothesis that as early as we have looked, the 5HT-cAMP-I KS cascade is in place and appears mature whereas the 5HT-PKC-I KV cascade does not emerge until much later. A similar temporal dissociation in the development of 5HT activation of PKA-and PKC-dependent pathways has been demonstrated for the development of 5HT responsiveness in the Retzius cell of the leech (26), suggesting that there may be a conserved function for the sequence of expression of PKA-and PKC-mediated pathways in development, perhaps in the regulation of outgrowth and neuronal morphogenesis (27).…”

“…3), or branching (23) of these neurons in lobsters. A wealth of supporting evidence in the literature suggests that serotonin can regulate the proliferation, growth, and connectivity of serotonin-sensitive neurons (22,(29)(30)(31)(32)(33)(34). In a striking example, serotonin has been shown to play a role in the development of the primary somatosensory map (28).…”
